The first part:Comparison of PI3 K,m TOR,p70S6 K protein and IFN-? expression in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage with normal puerperaObjective: To compare the expression of PI3 K,m TOR,p70S6 K protein and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage and normal puerpera.Method: 20 cases of HBV infected puerpera at immune tolerance stage and normal puerpera were selected,and p DCs of umbilical cord blood were isolated and cultured.Cp G-A was added on the 7th day.Cells and supernatant were collected after 24 hours,PI3 K,m TOR,p70S6 K protein were detected by Western blot;IFN-? in supernatant were measured by ELISA.Results: Compared with normal puerpera,PI3 K,m TOR,p70S6 K protein and IFN-? expression were reduced in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage,and the differences were statistically significant.The second part:The effects of Bushen Jiedu decotion on expression of PI3 K,m TOR,p70S6 K protein and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stageObjective:To study the effects of Bushen Jiedu decotion on expression of PI3 K,m TOR,p70S6 K protein and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage.Methods: 10 cases of HBV infected puerpera at immune tolerance stage were selected,and p DCs of umbilical cord blood were isolated and cultured.At the same time,the plasma containing drugs of Bushen decotion,Jiedu decotion and Bushen Jiedu decotion were prepared.On the 7th day,the p DCs were randomly divided into 5groups,Bushen drug plasma group,Jiedu drug plasma group,Bushen Jiedu drug plasma group,blank plasma group and blank group.After cultured with plasma for 48 hours,p DCs and supernatant were collected,PI3 K,m TOR,p70S6 K protein were detected by Western blot,IFN-? in supernatant were measured by ELISA.Results: The plasma containing drugs of Bushen Decoction and Bushen Jiedu Decoction can improve the expression of PI3 K,m TOR,p70S6 K protein,and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage,especially in the Bushen Jiedu group.Jiedu decotion can't improve the expression of PI3 K,m TOR,p70S6 K protein and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage statistically.Conclusion: 1.The expression of PI3 K,m TOR,p70S6 K protein and IFN-? in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage is reduced,and p DCs function is inhibited.2.Bushen Jiedu Decoction can significantly improve the expression of PI3 K,m TOR,p70S6 K protein in umbilical cord blood p DCs of HBV infected puerpera at immune tolerance stage,increase the secretion of IFN-? and improve the function of p DCs in patients infected with HBV.
