Mesenchymal stem cells(mesenchymal stem cells,MSCs)is the source of mesoderm have high ability to self-renewal and multi-directional differentiation potential of pluripotent stem cells,widely throughout a variety of organizations,can be in in vitro amplification,and under certain conditions to differentiate into osteoblasts,cartilage cells,fat cells,nerve cells and islet cells,etc.MSCs are pluripotent stem cells with the ability of "horizontal differentiation" or "cross-line differentiation",which not only support the growth of hematopoietic stem cells and repair acute injury with their own chemotactic properties,but also differentiate into a variety of tissue cells in vitro under different induction conditions.MSCs have a broad clinical application prospect,and are the first choice of seed cells for cell replacement therapy and tissue engineering,and are a research hotspot in the field of transplantation and the treatment of autoimmune diseases.This experiment in mice kidney mesenchymal stem cells(kidney mesenchymal stem cells,KMSCs)as experimental object,in vitro cultivation of cells separately after purification,biological characteristics and differentiation research,between tracer tag good mice kidney mesenchymal stem cells in the animal model of acute renal injury in mice,and to get the following results:1.Use ? KMSCs type collagen enzyme isolated from mice,under the condition of in vitro culture to 26,cell morphology of polygon and irregular shape at first,after three generations grow gradually unified spindle,colony growth.Rt-PCR showed that CD29,CD44,CD73 and CD105 expressed positive genes,and CD34 expressed negative genes.Immunohistochemical test results showed that KMSCs of mice positively expressed CD44,CD73,CD90 and CD105 surface markers.The number of KMSCs cells was recorded on the cell counting board of the hemocytometer,and the cell growth dynamics curve was plotted.The cell growth curve of KMSCs presented a typical "S" shape,which was basically consistent with the normal law of KMSCs proliferation in vitro.The results of population doubling time and clone formation ability of KMSCs showed that the cell proliferation efficiency before 5 generations was not as high as that between 5 and 20 generations,but with the increase of cell generation,the proliferation rate of KMSCs decreased and finally stagnated.In addition,studies have shown that KMSCs of mice can differentiate into osteoblasts,chondrocytes,adipocytes,hepatoid cells and neuron-like cells under specific induction conditions.Specific staining and rt-pcr identification have proved that KMSCs successfully differentiate into the above five types of cells.2.KMSCs of the mice labeled with the tracer were injected into the lps-induced acute renal injury model of the mice,and serum and kidney tissues were collected at different stages for relevant detection.After statistical analysis,the conclusion was drawn that KMSCs could indeed repair the acute renal injury of the mice.In summary,KMSCs of mice were successfully isolated and cultured in this experiment,and the immune repair function of KMSCs was verified in the animal model of acute renal injury of mice,providing experimental and theoretical basis for regenerative medicine and organ transplantation.
