In the research field of ethnic culture and medicine today,most of them are only studying one of them.Although there are also some documents that describe the relationship between the two,the characteristics,and points out the inadequacies of the full text.Objective:To establish a subcutaneous xenograft model of BALB/c nude mice transplanted with human hepatocellular carcinoma BEL-7402 cells to investigate the growth and angiogenesis effects of Tujia pip extract on transplanted tumors,and to use the P53/Gadd45a axis and its associated factor HIF-1?.NF-?B,STAT3,and VEGF are the entry points,and their effects on the mechanism of vascular endothelial growth factor(VEGF)in hepatocellular carcinoma are clarified.This will provide a new way for traditional Chinese medicine to treat liver cancer and lay a scientific foundation for the development and utilization of ethnic drugs.Methods:A subcutaneous transplantation model of human hepatocellular carcinoma BEL-7402 cells was established.After the completion of the modeling,30 tumor-bearing mice were randomly divided into control group,5-FU positive control group(5-FU),and sputum.The high-dose extract group(Fei Lian High,FLH),the limulus extract group(Fei Lian Medium,FLM),and the limulus extract low dose group(Fei Lian Low,FLL),6 in each group;Six normal nude mice with the same conditions as modeled nude mice were selected as the normal group.The Normal and Control groups were given normal saline 0.2 ml/time/d;the 5-FU group was given intraperitoneally with 3.0 ml/kg of 5-FU 0.2 ml/time/d,once every 2 days;FLH,FLM In the FLL group,0.24 g/kg,7.8 g/kg,and 5.2 g/kg of extract solution of Radix Paeoniae Rubra was administered 0.2 ml/time/d for 2 weeks.General index observations:1 Observe and record the general condition of nude mice every day.Measure the body mass every 2 days.Measure the volume of the transplanted tumor with the electronic vernier calipers.3 Calculate the tumor volume and tumor inhibition rate 3 After the completion of administration,obtain with the eyeball blood sampling method.The whole blood of nude mice was killed,then the mice were sacrificed by cervical dislocation.The livers of the mice were completely removed and the subcutaneously transplanted tumors were excised,and the morphology of the transplanted tumors was observed and compared.Histopathological observation:1 HE staining was used to observe the microstructure of the liver and transplanted tumor in nude mice;2 colorimetric method was used to detect the expression levels of liver function ALT,AST,GGT,GST in nude mice blood serum;3 SABC method was used to detect microvessel density in xenografts MVD;4 Immunohistochemical method was used to detect the positive expression of P53,Gadd45a,NF-?B,STAT3,p-STAT3,HIF-la and VEGF protein in the xenografts;5P53,Gadd45a and NF-?B were detected by Western blot.The expression levels of STAT3,p-STAT3,HIF-1a and VEGF proteins were detected.The expression of VEGF in nude mice serum was detected by ELISA.Results:General indicators:1 In terms of body mass of nude mice,body weight of nude mice in the Control group was significantly reduced compared with the Normal group(P<0.05).Compared with the Control group,body weight of the nude mice in the 5-FU group was significantly reduced,FLH,FLM The weight of nude mice in FLL group increased significantly(P<0.05).(2)Compared with the Control group,the transplanted tumor volume of the 5-FU group,the FLH group,the FLM group,and the FLL group significantly decreased(P<0.05).The tumor inhibition rates of tumor xenografts in each group were:5-FU group(43.52%),FLH group(35.15%),FLM group(19.91%),FLL group(16.45%).Histopathological indexes:1HE results showed that in the control group,the nude mice had large nests of cancer cells,and they showed small tumor necrosis and good cell morphology;in the 5-FU and FLH group,large cancer cells had large nests with large blood vessel necrosis and necrotic cells.In the FLM and FLL groups,there was only a small amount of tumor necrosis and necrotic cells.In the liver,normal,control,FLH,FLM,and FLL groups were normal in the liver of the nude mice,and inflammatory cells infiltrated in some areas of 5-FU.Edema and mild fibrosis.2 Colorimetric results showed that compared with the Normal group,the ALT,AST,GGT,and GST in the Control group were not significantly different(P>0.05);Compared with the Control group,the ALT,AST,GGT,and GST in the 5-FU group were significant.Increased(P<0.05).3SABC staining showed that MVD in 5-FU,FLH,FLM,and FLL groups was significantly lower than that in Control group(P<0.05).4 Immunohistochemistry results showed that P53,NF-?B,p-STAT3,HIF-1? and VEGF protein were strongly positive(+++)in the Control group and weakly positive in the 5-FU and FLH groups(Positively expressed((+++)in FLM and FLL groups.STAT3 protein was strongly positive in the control,5-FU,FLH,FLM,and FLL groups(+++).Gadd45a protein was weakly positive in the Control group.Expression(+)was strongly positive in the 5-FU and FLH groups(+++)and positive in the FLM and FLL groups(++).5 Western blot results showed that compared with the Control group,the expression of P53,NF-?B,P-STAT3,HIF-1a and VEGF protein in the 5-FU,FLH,FLM,and FLL xenografts in nude mice was significantly decreased(P<0.05),Gadd45a protein expression was significantly increased(P<0.05),and there was no significant difference in STAT3 protein expression(P>0.05).6 ELISA results showed that compared with the Normal group,the expression of VEGF protein in the nude mice serum of the Control group was significantly increased(P<0.05);compared with the Control group,the VEGF in the serum of the nude mice of the 5-FU,FLH,FLM,and FLL groups.Protein expression levels were significantly lower(P<0.05).Conclusion:(1)The Tujia medicine extract has the effect of increasing the body weight of BALB/c nude mice implanted in human hepatocellular carcinoma BEL-7402 cells,inhibiting the growth of subcutaneous xenograft tumors and the angiogenesis of xenograft tumors,and inhibiting the growth of liver cancer tumors.The mechanism may be achieved by inhibiting angiogenesis;(2)The nude mice subcutaneous xenograft tumor model does not have hepatotoxicity to the liver in a short term using Tujia pip extract;(3)Tujia drug extract can down-regulate the expression of P53,NF-?B,p-STAT3,HIF-la and VEGF in transplanted tumors of BALB/c nude mice implanted with human hepatocellular carcinoma BEL-7402 cells.Level;up-regulates Gadd45a protein expression levels;down-regulates VEGF protein expression levels in model serum.It was suggested that the extract of wolfberry may inhibit the angiogenesis of hepatocellular carcinoma by down-regulating the expression of VEGF protein through P53/Gadd45a axis.
