Objective: In this study,the imaging changes and the expression of important transcription factor runx-2 in the distraction site of tibial distraction model rats were studied,and the role of total flavones in distraction osteogenesis was investigated.Methods: 60 healthy SDF grade experimental rats were selected,and after 1 week of adaptive feeding,they were divided into model group,low dose group,medium dose group and high dose group by using random number table method.The model group was given normal saline by gavage after modeling,and the low-dose group was given flavonoids 67mg/kg by gavage.The medium dose group was administrated by intragastric administration of flavone 201mg/kg after osteotomy.The high-dose group was administrated by gavage of603mg/kg flavonoids after bone fragmentation after modeling.X-ray films were taken for 30 days after continuous administration of the drug in all experimental animals after modeling.After that,all rats were sacrificed,and the expression of Runx-2 in bone tissues was detected by Western-blot and RT-PCR.Data were collected for statistical analysis.Results: Compared with the model group,the imaging scores and Runx-2 gene expression were increased in the animal models with different doses of bone fragmentation supplementation(P<0.01).Imaging scores and Runx-2 gene expression were significantly higher in the medium and high dose groups than in the low dose group(P<0.01).The comparison between the medium dose group and the high dose group was not statistically significant(P>0.05).Conclusion: Osteopractic total flavone can up-regulate the expression of runx-2 protein gene in experimental animal models of tibial stretch osteogenesis,thereby improving osteogenesis,promoting bone mineralization,and accelerating fracture shaping.
