| Objective:To investigate the intervention effect and possible mechanism of weiwei tongtiao decoction in rats with precancerous lesions of atrophic gastritis.It provides laboratory basis for the clinical promotion of weiwei tongtiao decoction and promotes the inheritance and development of ethnic medicine theory.Methods:Ninety SD rats were randomly divided into normal group,model group,weifuchun control group,weiweitongdiao decoction high,medium and low dose groups,with 15 rats in each group.Rats in the normal group were routinely fed,and those in the experimental group were given N-methyl-N-nitro-N-nitrosoguanidine(MNNG)for free drinking,and supplemented with ranitidine hot saline and starvation and satiety disorder for the preparation of chronic atrophic gastritis precancerous lesion rat model.After the successful model preparation was confirmed by histopathological examination,the weiwei tongdiao decoction high(24g.kg-1/d),medium(12g.kg-1/d)and low dose(6g.kg-1/d)groups were given weiwei tongdiao decoction by gavage and the gastric fuchun control group was given gastric fuchun solution by gavage(0.45g.kg-1/d),once a day for 12 weeks.After treatment,the general conditions of rats in each experimental group were understood,H&E staining was used to observe the pathological changes of gastric mucosa in CAG precancerous lesion rats,the relative expression levels of IKK? and Bcl-2 mRNA in gastric mucosa were detected by qRT-PCR,and the expression levels of IKK? and Bcl-2 protein in gastric mucosa were detected by IHC.Spass24.0 was used to analyze the data results.Results:1.Effect of weiwei tongtiao decoction on general condition of CAG precancerous lesion rats:The rats in the model group showed slight body,dry and yellow fur,depilation,slow reaction,listlessness,tendency to curl and lie in a heap,or mental irritability,mutual biting,no desire to eat,thin and foul stool,and weight loss.Before treatment,the weight of the rats in the model group was significantly lower than that of the normal group(p<0.01),There was no significant difference in body weight in model groups(p>0.05);After treatment,the rats in each drug intervention group recovered,their fur glossiness improved,their diet increased,and their body weight increased significantly compared with the model group(p<0.01);There was no significant difference in body weight between the high-dose intervention group and the normal group(p>0.05)?2.Effect of weiwei tongtiao decoction on gastric mucosa of CAG precancerous lesion rats:In the model group,intrinsic gland atrophy and mild to severe intestinal metaplasia were observed under gastric mucosa microscopy,and epithelial cell hyperplasia and disorganized cell arrangement were observed in some tissues,with poloidal disappearance and large and deeply stained nuclei.After treatment,the degree of gastric mucosal lesions in rats in the weiwei tongdiao decoction high,medium and low intervention groups was significantly improved,especially in the weiwei tongdiao decoction high dose intervention group.3.Effect of weiwei tongtiao decoction on the expression of IKK? and Bcl-2 in gastric mucosa of experimental rats:The expression levels of IKK?,Bcl-2 mRNA and protein in gastric mucosa of the rats in each drug intervention group were significantly lower than those in the model group(p<0.05);Compared with the normal group,the expression levels of IKK?,Bcl-2 mRNA and protein in the high-dose intervention group of weiweitongtiao decoction showed no statistical difference(p>0.05),The expression levels of IKK?,Bcl-2 mRNA and protein in weiwei tongtiao decoction were significantly increased in the medium and low dose intervention groups and weifuchun control group(p<0.01),The expression levels of IKK?,Bcl-2 mRNA and protein in gastric mucosa in the low-dose intervention group of weiweitongtiao decoction were similar to those in the weifuchun control group(p>0.05).Conclusion:1.Weiwei tongdiao decoction can block,improve or even partially reverse gastric mucosa atrophy,intestinal metaplasia and low-grade intraepithelial neoplasia in CAG precancerous lesion rats,among which weiwei tongdiao decoction high-dose intervention group was superior to other drug intervention groups.2.The intervention mechanism of weiwei tongtiao decoction on CAG precancerous lesion may be related to the down-regulation of IKK?,Bcl-2 mRNA and protein expression levels in gastric mucosa. |
