1 Objective:Skeletal muscle injury is one of the common injuries in sports injuries.Although skeletal muscle has the ability to repair itself,excessive damage will lead to functional degradation of skeletal muscle.Studies have found that acupuncture can reduce skeletal muscle damage when skeletal muscle is injured,but the specific mechanism is not yet clear.In addition,studies have found that acupuncture is closely related to calpains.Therefore,this experiment aims to investigate the effect of calpains on skeletal muscle injury after acupuncture.2 Methods:After one week of acclimation,the rats were randomly divided into 4 groups using random grouping method.Quiet group?C?,exercise group?E?,exercise acupuncture group?EA?,exercise acupuncture plus TRP channel blocker group?EAI?.The quiet group?C?does not intervene.Exercise group?E?,exercise plus acupuncture group?EA?,exercise acupuncture plus blocker group?EAI?performed 90 min eccentric exercise with the same load.This centrifugal exercise is a downhill running exercise,the treadmill slope is-16°,the speed is 16 m·min-1,the exercise time of each group is 5 minutes,a total of 18 groups,and the rest between the groups is 2 minutes.In the EA group,only one oblique puncture ?diagonal puncture angle of 300?was performed on the femoral middle muscle of the rats.When obliquely puncturing the middle femoral muscle,the time to retain the needle is 4min.During this period,no movements such as pinching and lifting can be done.After completing the exercise,the EAI group was injected with 0.2%Gdcl3 salt solution immediately,and then performed a diagonal puncture?diagonal puncture angle 300?of the femoral middle muscle of rats.In the end,each group took the mid-femoral muscles of rats'lower limbs at 1h,48h and 120h.The left lower extremity middle femoral muscle was observed by SDS polyacrylamide gel electrophoresis to observe protein activity,and the right lower extremity middle femoral muscle was observed with transmission electron microscope to observe the ultrastructural Z-line of skeletal muscle.3 Results:3.1 The effect of acupuncture intervention on the change of calpains:?1?Compared with C:E showed an increasing trend compared with calpain-1 and calpain-3 in group C,both significantly increased with group C?P<0.05?,and calpain-2 was very significant Sexual increase?P<0.01?.EA was significantly higher than that in group C?P<0.05?.EAI was significantly higher than that of group C?P<0.05?;?2?Comparison of different groups at the same time:EA group showed a downward trend compared to group E,calpain-1,calpain-2,and calpain-3,which were all significantly different from group E Difference P<0.05;calpain-1,calpain-2 and calpain-3 in the EAI group showed an upward trend compared with the EA group.There were significant differences between the EA group and P<0.05.?3?Compared with the same group at different times:the E group of calpain-1 and 3reached the peak value at 48h,when 1h and 120h were compared with 48h respectively,P<0.05 was significantly different.The calpain-2 E group reached the peak at 48h,P<0.01when 1h and 120h were compared with 48h respectively,and calpain-2 was very significant difference;the EA group of calpain-1 reached the peak at 48h,1h and 120h were respectively 48h Compared with calpain-1,P<0.01 was a very significant difference and P<0.05 was a significant difference.The EA group of calpains-2 and-3 reached the peak at120h,P<0.05 at 1h and 48h compared with 120h,respectively,which were very significant differences;the EAI group of calpain-1,2,and 3 arrangement:Z-line integrity,Z-line breakage,and Z-line fuzzy changes.?1?Compared with group C,the completeness of P<0.01Z line is significantly reduced,the breakage of P<0.01Z line is extremely significantly increased,48h P<0.01Z line blur change is significantly increased;?2?Comparison of different groups at the same time phase,1h When the ratio of E to EA group was P<0.05,the integrity of Z line was significantly reduced,and the fracture and ambiguity of Z line were significantly increased.Compared with the EA group,EAI P<0.05,the Z line integrity is significantly reduced,and the Z line fracture and ambiguity are significantly increased;at 48h,the E and EA group is P<0.05,the Z line integrity is significantly reduced,and the Z line crack Compared with the ambiguity,EAI and EA group P<0.05,the Z-line integrity is significantly reduced,and the Z-line crack and ambiguity are significantly increased.At 120h,the E and EA groups had a P<0.05,and the Z line integrity was significantly reduced.The EAI and EA groups were P<0.05,and the Z line integrity was significantly reduced.Therefore,the centrifugal motion destroys the integrity of the Z line and increases the ambiguity and fracture of the Z line.Acupuncture can restore the ambiguity and breakage of the Z line.?3?Compared with different time of the same group,compared with EA,1h,48h,120h P<0.01Z line integrity is significantly increased,48h P<0.01 fracture blur is significantly reduced;EA group is compared with EAI group,P The integrity of P<0.01 is very significantly increased,and the fracture of P<0.01 is very significantly reduced at 48h4 Conclusion:4.1 Acute eccentric exercise caused an increase in the activity of skeletal muscle cells calpain-1,2,and 3,causing skeletal muscle ultrastructural damage.4.2 Acupuncture down-regulated the activity of calpain-1,2,3 of skeletal muscle in acute eccentric exercise,and reduced the degree of sarcomere structure damage.Its effect mechanism is related to the activity of TRP channel.
