Objective:To explore the mechanism of different concentrations of Lipi compound preparation in treating anorexia rats,and to understand its effects on gastrointestinal motility,hypothalamus and serum motilin,gastrin,somatostatin and neurotensin.The clinical application of the agent provides experimental basis.Methods:After adaptive feeding for 96 days,96 rats were randomly divided into blank group 12(referred to as group A)and model group(group B),group A normal feed,group B special feed 14 day.The model group used the etiological simulation method to establish anorexia model rats.After successful modeling,group B was randomly divided into female saline group(referred to as group C),spleen compound preparation low dose group(referred to as group D),and spleen compound.The drug preparation group(referred to as E group),the spleen compound preparation high-dose group(referred to as F group),the zinc sulfate group(referred to as G group),the multomorphine group(referred to as H group)a total of 8 groups,each group of rats 12 only.Group A was a blank control group,group B was a model control group,group C was treated with normal saline,group D,E,and F were treated with low,medium,and high doses of spleen compound preparation,and group G was treated with zinc sulfate.The group H was treated with multomorphine,once a day,after 7 consecutive days.Gastric emptying rate and small intestine propulsion rate were measured by carbon perfusion method.Serum and hypothalamic motilin and serum gastrin were measured by ELLSA method.Plasma neurotensin and somatostatin were determined by radioimmunoassay.The experimental results were statistically processed using SPSS 23.0 statistical software.The measurement data were consistent with the normal distribution and the mean ± standard deviation(X±S).The comparison between groups was performed by one-way ANOVA.When the variance was equal,the LSD test was selected.When the variance was not uniform,Dunnett's T3 method was used to analyze,and P<0.05 was considered statistically significant.Results:1.Compared with the normal group,the model group and the normal group showed significant differences in food intake and body weight(P<0.05),indicating successful modeling.2.After the intervention,the food intake of the anorexia rats was significantly different from that of the model group.The zinc sulfate group was followed by the saline group and the Lipi spleen compound preparation medium dose group.After the intervention,the body mass of the anorexia rats in each treatment group was up-regulated and the model group.There were differences,followed by zinc sulfate group,normal saline group,the Lipi spleen compound preparation medium,high and low dose group,and morpholine group.3.Gastric emptying rate and small intestine propulsion rate:After model establishment,the gastric emptying rate and small intestinal propulsion rate of the anorexia rats in the model group were all down-regulated(P<0.05).Except the saline group,there were no difference in the small intestine propulsion rate after the intervention treatment(P>0.05),the treatment group had different effects on gastric emptying rate and small intestine propulsion rate(P<0.05).The Lipi spleen compound preparation had the best effect in the middle dose group,and the morpholine group was superior to the zinc sulfate group.4.Gastrointestinal hormones:After model establishment,there were significant differences in hypothalamic and peripheral blood motilin and gastrin in the model group of anorexia rats(P<0.01),and neurotensin up-regulation was different(P<0.05).There was a significant difference in up-regulation of somatostatin(P<0.01).After the intervention,the middle dose group of Lipi compound preparation can significantly increase the content of motilin in hypothalamus and peripheral blood of anorexia rats,reduce neurotensin and somatostatin,and the low dose group of Lipi compound preparation can improve the gastrin content.In the middle dose group,except for the hypothalamic motilin,there was no difference in the zinc sulfate group(P>0.05).The morphine group and the zinc sulfate group could increase the motilin and gastrin levels to different degrees and reduce the nerve drop.Pressure hormone,somatostatin content.Conclusion:The compound preparation of spleen and spleen is effective in treating anorexia The mechanism may be to increase the content of motilin and gastrin in anorexia rats,reduce the content of neurotensin and somatostatin,and promote the increase of feeding behavior and promote gastric emptying in anorexia rats.Rate,small intestine propulsion rate,enhance gastrointestinal motility and restore food intake,increase body mass,cure anorexia.The medium dose is the best in different concentrations.This dose is used for the treatment of anorexia doses.It provides experimental theoretical basis for clinical use,and lays a foundation for promoting the application of famous Chinese medicine practitioners.It also lays a foundation for the promotion of new dosage forms.
