Objective: To explore the effect of Lithospermum erythrorhizon on the prevention of pericardial adhesion after cardiac surgery in rabbits and its possible mechanism.Materials and methods: 32 Japanese white rabbits were selected and randomly divided into four groups according to the random number table method: the blank control group,the model control group,the Gel for Prevention of Postsurgical Adhesion group and the Lithospermum erythrorhizon group,with 8 male rabbits in each group.The pericardium of each group was exposed by median sternotomy,and then the pericardium was opened.In the blank control group,there was no external interference such as swab wiping and blood injection.The other groups repeatedly wiped the parietal and visceral pericardium of the fixed area at the bottom of the heart with sterile small swab for 100 times,and injected autogenous venous blood into the pericardium cavity,establishing pericardial adhesion model.In the model control group,the pericardium was opened and the model was established,but no drugs were infused into the pericardium;in the Gel for Prevention of Postsurgical Adhesion group,the Gel for Prevention of Postsurgical Adhesion was infused into the pericardium;in the Lithospermum erythrorhizon group,Lithospermum erythrorhizon solution was infused into the pericardium;.30 days after the operation,the rabbits were killed by air embolism.The degree of pericardial adhesion and thickening were observed.The results of ELISA for fibronectin(FN)? integrin(ITG)? focal adhesion kinase(FAK)in the pericardial tissues of rabbits were determined by ELISA,and the section staining of pericardial tissue was analyzed by light microscopy.Results: 1.Pathological observation and rating results show that the model control group is significantly thicker than the blank control group,the Sebital group and the comfrey group.The pericardium cannot see the blood vessels or myocardium on the surface of the heart through the pericardium.The adhesion is obvious and requires blunt separation.The four groups had significant differences in the degree of pericardial adhesion(F[3,28]=41.797,P<0.05).The score of the blank control group was lower than that of the model control group,with a significant statistical difference(MD=-2.38,P<0.05).);The score of the model control group is higher than that of the Sebital group(MD =-1.75,P<0.05);The score of the model control group is higher than the comfrey group(MD=1.88,P<0.05));The comfrey group score is lower than that of the Sebital group,no statistical significance(MD=-0.13,P>0.05).The four groups had significant differences in the degree of pericardial thickening(F[3,28]=69.395,P<0.05).The score of the blank control group was lower than that of the model control group,with a significant statistical difference(MD=-2.38,P< 0.05);the points of the model control group are higher than those of the Sebital group,and there are significant differences(MD=1.75,P<0.05);the points of the model control group are higher than the comfrey group,and there are significant differences(MD=1.75,P<0.05);The average and variance of the points in the comfrey group and the Sebital group are equal and have no statistical significance.2.The observation of tissue section showed that there was a significant difference between the blank control group,the Gel for Prevention of Postsurgical Adhesion group as well as the Lithospermum erythrorhizon group compared and the model control group.The myocardium and pericardium in the tissue section of the model control group had obvious inflammatory cell infiltration,and the lymphocyte and plasmocyte increased significantly.However,there was basically no such phenomenon in the blank control group;in the other two groups,there was only a small amount of infiltration,but no significant increase in lymphocytes and plasmocytes.3.Tissue ELISA test results showed that: in the results of Fn,In the results of Fn,there were significant differences between the four groups(F[3,28]=82.621,P<0.05);the count of the blank control group was lower than that of the model control group,and there was a statistically significant difference(MD=-8.23,P< 0.05);the count of the blank control group was lower than that of the Sebital group,and there was no statistical difference(MD=-0.65,P> 0.05);the count of the blank control group was lower than that of the comfrey group,and there was no statistical difference(MD=-1.06,P> 0.05);the count of the model control group is higher than that of the Sebital group,and there is a significant difference(MD=7.58,P<0.05);the count of the model control group is higher than the comfrey group,and there is a significant difference(MD=7.17,P <0.05);the count of the comfrey group was higher than that of the Sebital group,and there was no statistical difference(MD=0.41,P> 0.05).In the results of ITG,the four groups have significant differences(F [3,28]=43.330,P<0.05);the blank control group and the model control group have significant statistical differences(MD=-43.75,P<0.05);blank The count of the control group was lower than that of the Sebital group,and there was no statistical difference(MD=-13.48,P> 0.05);the count of the blank control group was lower than that of the comfrey group,and there was no statistical difference(MD=-12.61,P> 0.05);The model control group has a higher count than the Sepital group,and there is a significant difference(MD=30.27,P<0.05);the model control group has a higher count than the comfrey group,and there is a significant difference(MD=31.14,P <0.05);the count of the comfrey group was lower than that of the Sebital group,and there was no statistical significance(MD=-0.86,P> 0.05).In the FAK results,there were significant differences between the four groups(F[3,28]=112.132,P<0.05);the count of the blank control group was lower than that of the model control group,and there was a statistically significant difference(MD=-5.89,P< 0.05);the count of the blank control group was lower than that of the Sebital group,and there was no statistical difference(MD=-0.55,P> 0.05);the count of the blank control group was lower than that of the comfrey group,and there was no statistical difference(MD=-0.46,P> 0.05);the count of the model control group is higher than that of the Sebital group,and there is a significant difference(MD=5.34,P<0.05);the count of the model control group is higher than the comfrey group,and there is a significant difference(MD=5.43,P<0.05);the comfrey group count was higher than that of the Sepital group,and there was no statistical significance(MD=0.09,P> 0.05).Conclusion: 1.Lithospermum can effectively prevent pericardial adhesion after rabbit cardiac surgery,and has a deeper research value.2.Lithospermum erythrorhizon may reduce the concentration of fibrin in extracellular matrix and the incidence of pericardial adhesion by inhibiting the signal transduction and expression for FN-ITG-FAK signaling transduction.
