Purpose:Salt sensitive hypertension is a kind of essential hypertension,which is a special type of hypertension disease with significantly increased blood pressure after high salt intake.Acupuncture at "Sanyinjiao" point in traditional Chinese medicine has a good therapeutic effect on it.So in this experiment,Dahl salt sensitive rats were selected as the animal model of salt sensitive hypertension,Dahl salt resistant rats were used as the normal control group to observe,the method of electroacupuncture stimulating "Sanyinjiao" point was used to observe the control of Electroacupuncture on the blood pressure of salt sensitive hypertensive rats,the expression of Rac1 MR signal pathway was detected by fluorescence quantitative PCR and WB technology,and ELISA method was used To detect the expression of aldosterone in plasma in order to analyze whether electroacupuncture at "Sanyinjiao" point has a certain regulatory effect on salt sensitive hypertension.To provide some experimental basis for the treatment of salt sensitive hypertension.Materials and methods: 10 6-week-old salt resistant rats(DR)and 30 6-week-old salt sensitive rats(DS)were randomly divided into model group,electroacupuncture group and non acupoint group.At the time of purchase,the rats were 5 weeks old and their weight was200-220 g.After blood pressure measurement,the rats were fed with conventional feed for 1week,and then fed with high salt feed of 8% Na Cl.After the rats reached the level of hypertension(? 140 / 90 mm Hg),the rats were considered to have met the model standard.The rats were stopped feeding and began to receive electroacupuncture treatment.Rats in the normal group and the model group were only fixed on the barbed wire every day without acupuncture;rats in the acupuncture group were fixed in the same way as those in the normal group and the model group,and were treated with electroacupuncture.The "Sanyinjiao" points on both sides of the rats were taken for acupuncture treatment,with a depth of about5 mm,and the sd-ii electroacupuncture therapeutic apparatus was connected for electroacupuncture stimulation treatment.Intensity: rats were treated with electroacupuncture The limb tremor is degree(1-2ma),stimulation frequency: density wave length,density wave4 Hz,density wave 20 Hz.The period of density wave was 6S,each time for 20 min,once a day for two weeks.The rats in the non meridians and non acupoints group were fixed by the same method as the normal group and the model group,and a fixed control point was selected to be the non meridians and non acupoints [1] within the 10-15 mm of bilateral iliac crest and 20 mm of the posterior midline side opening of the rats.The depth of acupuncture was 5 mm,and the electric acupuncture was connected to stimulate the rats.At the end of the treatment,the plasma of abdominal aorta was anesthetized,and the content of aldosterone in the plasma was measured by ELISA.The expression of Ras related C3 botulinum toxin substrate 1(Rac1)and salt glucocorticoid receptor(MR)m RNA was measured by RT-PCR.The expression of Ras related C3 botulinum toxin substrate 1(Rac1)and salt were measured by WB Protein expression of corticosteroid receptor(MR).The experimental data were analyzed by spss19.0,and the results were expressed by mean ± standard deviation(x ± s).When the data between groups need to be compared,one-way ANOVA is used.When the variance between groups is the same,LSD test is used.When the variance between groups is not the same,tamhane's T2(m)method is used.When p < 0.05,the difference was statistically significant.Results:1.Changes of blood pressure in each group With the beginning of acupuncture treatment,the blood pressure of the acupuncture group began to decrease gradually,and gradually approached the normal group;although the blood pressure of the non acupoint group decreased,the curative effect was obviously not obvious in the acupuncture group.2.Expression of Rac1 m RNA and protein in adrenal cortex of rats in each group According to the statistical results:The m RNA content of Rac1 in the adrenal cortex of the non meridians and non meridians group was higher than that of the normal group(P < 0.05),and the m RNA content of Rac1 in the adrenal cortex of the acupuncture group was lower than that of the model group and non meridians and non meridians group(P < 0.05).There was no significant change in the m RNAcontent of Rac1 in the adrenal cortex of rats in the non meridian non acupoint group compared with that in the model group(P < 0.05).The protein content of Rac1 in the adrenal cortex of rats in the non meridian non acupoint group and the model group was higher than that in the normal group(P < 0.05),and the protein content of Rac1 in the adrenal cortex of rats in the acupuncture group was lower than that in the model group and the non meridian non acupoint group(P < 0.05).There was no significant difference in the protein content of Rac1 in the adrenal cortex between the non acupoint group and the model group(P < 0.05).The m RNA content of MR in the adrenal cortex of the rats in the non meridian non acupoint group and the model group was higher than that in the normal group(P < 0.05),and the m RNA content of MR in the adrenal cortex of the rats in the acupuncture group was lower than that in the model group and the non meridian non acupoint group(P < 0.05).There was no significant change of Mr m RNA content in the adrenal cortex of rats in the non meridian non acupoint group compared with the model group(P < 0.05).The protein content of MR in the adrenal cortex of the rats in the non meridian non acupoint group and the model group was higher than that in the normal group(P < 0.05).The protein content of MR in the adrenal cortex of the rats in the acupuncture group was lower than that in the model group and the non meridian non acupoint group(P < 0.05).Compared with the model group,the protein content of MR in the adrenal cortex of the non meridian non acupoint group had no significant change(P < 0.05).3.Expression of aldosterone in plasma of rats in each group The content of aldosterone in the plasma of rats in the non acupoint group and the model group was higher than that in the normal group(P < 0.05),and the content of aldosterone in the plasma of rats in the acupuncture group was lower than that in the model group and the non acupoint group(P < 0.05).Compared with the model group,the content of aldosterone in the non acupoint group had no significant change(P < 0.05)Conclusion:1.electro acupuncture at "Sanyinjiao" can effectively regulate the Rac1-MR pathway,which can reduce the content of Rac1 and MR m RNA and protein content in the adrenal cortex of rats,thereby reducing the reabsorption of sodium ions by the kidneys and reducing the retention of water and sodium in the kidneys,thereby reducing the damage of high salt intake to the kidneys and regulating the hypertension induced by high salt intake.2.electro acupuncture at "Sanyinjiao" can effectively reduce the content of aldosterone in plasma,and then reduce the reabsorption of sodium ions by the kidneys,reduce the retention of water and sodium in the kidney,thereby playing a regulatory role in blood pressure.
