| Purpose:To observe the effects of total flavonoids and total saponins of Huangqigegen Decoction and its compatibility on glucose and lipid metabolism and inflammatory response in the liver of diabetic rats,and explore its mechanism of action.Material and method:The 86 SPF SD rats(male)were divided into 6 groups as experimental subjects.11 rats in normal group(Nor),15 in model group(a1b1),15 in jin-qi-jiang-tang-table grop(Jinqi),15 in total flavonoids group(a2b1),15 in total saponins group(a1b2),15 in total flaonoids and saponins group(a2b2).Experimental rats(except the normal group)were induced by a one-time intraperitoneal injection of STZ(46mg·kg-1)for diabetic model.Molded manufacturing on the same day as intragastric administration.The dose is 10 m L·kg-1·d-1in Nor and a1b1 group,Jinqi group is 1.47g·kg-1·d-1,a2b1 group is0.106g·kg-1·d-1,a1b2 group is 0.074g·kg-1·d-1,a2b2 group is 0.180g·kg-1·d-1.Rats in each group continued to administer for 30 days.This study mainly tests the blood glucose and blood lipids of experimental rats.Real-time PCR was used to detect the expression of Insulin receptor substrate-2(IRS-2),Phosphatidylinositol-3-kinase(PI3K),Protein kinase B-2(Akt2),Glucose transporter-4(GLUT-4),Glycogen synthase kinase 3β(Gsk-3β),Adiponectin receptors-1(Adipo R1),AMP-activated protein kinase(AMPK),P38Mitogen-activated protein kinase(P38MAPK),Peroxisome proliferator-activated receptor gamma(PPARγ),Liver X receptor α(LXRα),Sterol regulatory element binding proteins 1(SREBP1)mRNA in liver tissue.Statistical Product and Service Solutions 17.0 is used data processing.Results:1.Effect on blood glucose of experimental rats: Before modeling,there was no significant difference in blood glucose among rats in each group(P>0.05).After modeling,the blood glucose of rats in a1b1 group was significantly increased,which has statistically significant(P<0.05).On the 7th day of modeling,there was no significant difference in blood glucose between the drug group and the a1b1 group(P>0.05);on the 30 th day of modeling,the blood glucose of the a1b1 group was significantly increased;compared with the a1b1 group,the blood glucose was significantly reduced in the a2b1 group and a1b2 group,which has statisticallysignificant(P<0.05),and there was no interaction between total flavonoids and total saponins(P=0.064>0.05).2.Effects on blood lipid in experimental rats: Compared with the Nor group,the levels of CHO and TG in the a1b1 group were significantly increased(P<0.05).Compared with the a1b1 group,total flavonoids could significantly reduce the CHO concentration.The effect of total saponin was not significant,and there was no interaction between total flavonoids and total saponins(P>0.05);total flavonoids and total saponins could reduce TG levels,and there was an interaction between them,but the single use of herb is better than the combination.3.Effects on the mRNA expression of IRS-2,PI3 K,Akt2,Gsk-3β and GLUT-4 in experimental rats:(1)Effects on the mRNA expression of IRS-2 in the liver tissue of experimental rats:Compared with the Nor group,the expression of IRS-2 mRNA in a1b1 group was significantly decreased.total saponins can effectively enhance IRS-2 mRNA expression.The effect of total flavonoids is not significant,and there is an interaction between them,but combination was not as good as a single drug.(2)Effects on the mRNA expression of PI3 K in the liver tissue of experimental rats:Compared with the Nor group,the expression of PI3 K mRNA in a1b1 group was significantly decreased.The total flavonoids and total saponins could promote the expression of PI3 K mRNA.There was no interaction between them(P>0.05).(3)Effects on the mRNA expression of Akt2 in the liver tissue of experimental rats:Compared with the Nor group,the expression of Akt2 mRNA in a1b1 group was significantly decreased.The total flavonoids and total saponins could promote the expression of Akt2 mRNA.but combination was not as good as a single drug.(4)Effects on the mRNA expression of Gsk-3β in the liver tissue of experimental rats:Compared with the Nor group,the expression of Gsk-3β mRNA in a1b1 group was significantly rise.Total flavonoids and total saponins could reduce the expression of Gsk-3β.The interaction between them was obvious,but total flavonoid effect is better.(5)Effects on the mRNA expression of GLUT-4 in the liver tissue of experimental rats:Compared with the Nor group,the expression of GLUT-4 mRNA in a1b1 group was significantly decreased.And total flavonoids could effectively increase the expression ofGLUT-4 mRNA,The effect of total saponins is not significant,There was an interaction between total flavonoids and total saponins.But the effect of total flavonoids alone is more prominent.4 Effects on the expression of Adipo R1,AMPK,P38 MAPK,PPARγ,LXRα and SREBP1 mRNA in liver tissue:(1)Effects on the mRNA expression of Adipo R1 in the liver tissue of experimental rats:Compared with the Nor group,the expression of Adipo R1 mRNA in a1b1 group was significantly decreased.total flavonoids and total saponins could effectively promote Adipo R1 mRNA expression,and the interaction between them was obvious,but it was better to use single drug.(2)Effects on the mRNA expression of AMPK in the liver tissue of experimental rats:Compared with the Nor group,the expression of AMPK mRNA in a1b1 group was significantly decreased.and total flavonoids could significantly enhance the expression of AMPK mRNA.The effect of total saponin was not obvious,and the Compatibility between them was not better than that of the total flavonoids.(3)Effects on the mRNA expression of P38 MAPK in the liver tissue of experimental rats:Compared with the Nor group,the expression of P38 MAPK mRNA in a1b1 group was significantly rise.Total flavonoids and total saponins could decrease the expression of P38 MAPK mRNA.There was an interaction between total flavonoids and total saponins.But the total saponins alone was better.(4)Effects on the mRNA expression of PPARγ in the liver tissue of experimental rats:Compared with the Nor group,the expression of PPARγ mRNA in a1b1 group was significantly decreased.and total flavonoids could effectively enhance the expression of PPARγ mRNA.Total saponins and the interaction between total flavonoids and total saponins was not obvious.(5)Effects on the mRNA expression of LXRα in the liver tissue of experimental rats:Compared with the Nor group,the expression of LXRα mRNA in a1b1 group was significantly decreased.and total flavonoids and total saponins could effectively promote the expression of LXRα mRNA.The interaction between them was not obvious.(6)Effects on the mRNA expression of SREBP1 in the liver tissue of experimental rats:Compared with the Nor group,the expression of SREBP1 mRNA in a1b1 group was significantly rise.The total flavonoids and total saponins could effectively reduce the level of SREBP1 mRNA.The interaction between them was obvious,but the combination was not as good as single drug.Conclusion:1.The total flavonoids,total saponins and their compatibility in Huangqi Gegen Decoction can play an effective hypoglycemic effect.In this hypoglycemic process,the Compatibility between total flavonoids and total saponins exert their own characteristics.Total saponins are mainly involved to the expression of IRS-2 mRNA in the upstream of insulin signaling pathway.The main role of total flavonoids in the downstream of insulin signaling pathway is closely related to the expression of GLUT-4 and Gsk-3β mRNA.The hypoglycemic mechanism of Compatibility between them is mainly involved to the PI3K/Akt signaling pathway.2.The total flavonoids,total saponins and their compatibility from Huangqi Gegen Decoction can effectively reduce lipids,but the mechanism of action is slightly different.Total flavonoids may play a major role in regulating AMPK and PPARγ mRNA gene expression;The regulatory effect of total saponins on P38 MAPK mRNA is more significant;the effects between them may be related to the activation of the AMPK/SREBP1 signaling pathway,and further to play a series of cascades in p38 MAPK,PPARγ,LXRα and other signaling factors.3.Total flavonoids,total saponins and their compatibility from Huangqi Gegen Decoction can play an effective anti-inflammatory effect,which is mainly related to AMPK、P38MAPK、PPARγ、LXRα、SREBP1 signal factors. |
