Purpose: Observing whether the femoral head recovery tablet can promote the ulna bone defect in rabbits,and its effect on the expression of serum PINP,tracp-5b and bmp-2 in bone defect.To explore the mechanism of femoral head rehabilitation tablets in promoting bone defect repair.Materail and Method:36 rabbits were numbered and a bone defect model of the left ulna middle segment was made.The defect length was 15 mm and periosteal membrane was not retained.After making the model,32 rabbits were randomly divided into four groups by computer: traditional Chinese medicine group(group 2),western medicine group(group 3),combined drug group(group 4)and blank group(group 1),with 8 rabbits in each group.Chinese medicine group was given 2ml/kg by intragastric administration of Gutou Kangfu tablet every day.Western medicine group was given2ml/kg by intragastric administration of Ossotide tablet every day.In the combined treatment group,2ml/kg by intragastric administration of Gutou Kangfu tablet and osteopeptide tablet were given every day.The blank group was given 2ml/kg by intragastric administration of normal saline every day.Elisa was used to detect the expression of serum PINP and tracp-5b before modeling and 6 ? 12 weeks after modeling.At each time node of 6 and 12 weeks after modeling,the method of random number table was used to extract 4 rabbits from 4 groups,and then the complete ulna specimens were obtained.Micro-ct was used to detect the healing of bone defects in each group.The three-point bending experiment was carried out with biomechanical instrument to detect the maximum load of regenerated new bone and to compare the groups.He staining was used to observe the microscopic condition of local bone defect repair.The expression of BMP-2 in bone defect was detected by Western Blot.Results:1 Elisa results of serum PINP showed that there were no significant difference in serum PINP in each groups before the model was established(p>0.05),and the baseline was comparable.The PINP of 6 weeks after modeling: the combination group > TCM group > Western medicine group > blank group,the difference between the drug intervention group and the blank group was statistically significant(p<0.05),and the two drugs had a synergistic effect(p=0.000).The PINP of 12 weeks after modeling: the combination group > TCM group > Western medicine group > blank group,the difference between the combination group and the blank group was statistically significant(p<0.05),but there were no synergistic effect between the two drugs(p=0.658).2 Elisa results of serum TRACP-5b showed that there was no statistically significant difference in serum TRACP-5b in each group before modeling(p > 0.05),and the baseline was comparable.The TRACP-5b of 6 weeks after modeling: the combination group < Western medicine group < TCM group < blank group,the difference between the drug intervention group and the blank group was statistically significant(p<0.05),and the two drugs had a synergistic effect(p=0.006).The TRACP-5b of 12 weeks after modeling: the combination group < TCM group < Western medicine group < blank group,the difference between the drug intervention group and the blank group was statistically significant(p<0.05),and the two drugs had a synergistic effect(p=0.018).3 The maximum load /N of radius and ulna was shown : 6 weeks after modeling:combination group > western medicine group > TCM group > blank group,the difference between the drug intervention group and the blank group was statistically significant(p < 0.05),and the two drugs had synergistic effect(p=0.006).12 weeks after modeling: combination group > TCM group > Western medicine group > blank group,the difference between the drug intervention group and the blank group was statistically significant(p < 0.05),and the two drugs had synergistic effect(p=0.000).4 Micro-ct examination of bone defect healing showed that the radial bone cortex of the Chinese medicine group and the western medicine group had been connected in 6 weeks,and there was a gap in the ulnar side,and there was basically no difference between the two groups.In 12 weeks,the ulnar gap between the two groups was further reduced,and there was basically no difference between the two groups.Combination group in 6weeks,the medullary cavity was gradually formed and the ulnar cortical bone began to connect.In 12 weeks,the medullary cavity was basically recalculated and the ulnar cortex was connected,but the newly formed bone density was slightly lower than the surrounding normal bone.The blank group in 6 weeks,was little bone growth in the radialis of the blank group,but the two ends were not connected.In 12 weeks,there was a little bone growth in the radial side compared with before,but the two ends of the bone were not connected.5 HE staining showed that the trabecular arrangement at the broken end was disordered in the blank group at 6 and 12 weeks,there was more fibrous tissue at the broken end and a small amount of chondrocytes.In 6 weeks,there were more trabeculae in the Chinese medicine group and the western medicine group.In 12 weeks,trabeculae were further increased and began to be connected to each other.Trabeculae began to connect with each other at 6 weeks in the combination group,and more chondrocytes appeared.Trabeculae density further increased at 12 weeks.6 Local expression of bmp-2 in bone defect:Bmp-2 gray value 6 weeks after modeling,combination group > Western medicine group > TCM group > blank group,the difference between the drug intervention group and the blank group was statistically significant(p<0.05).However,there was no synergistic effect between the two drugs(p=0.569).Bmp-2 gray value 12 weeks after modeling,combination group > Western medicine group > TCM group > blank group,the difference between the drug intervention group and the blank group was statistically significant(p<0.05),and the two drugs had a synergistic effect(p=0.000).Conclusion:1 Gutou Kangfu tablet can promote the healing of bone defects.2 Gutou Kangfu tablet can promote bone defect repair by increasing the expression of PINP and bmp-2 and inhibiting the expression of tracp-5b.3 There is a synergistic effect between Gutou Kangfu tablet and Ossotide tablet.
