Purpose:1?The contents of flavonoids in Platycladus orientalis leaves were determined by high performance liquid chromatography(HPLC)with seven flavonoid components,kaempferol,myricetrin,quercitrin,myricetin,quercetin,hinokiflavone and amentoflavone as indicators.2?The content of flavonoids in Platycladus orientalis leaves and processed Platycladus orientalis leaves was determined by HPLC.And compare the content changes of 7 components before and after processing.3?The content of 18 amino acids such as aspartic acid in the leaves of Platycladus orientalis from Guiyang,Guizhou Province,was determined by dual-pump high performance liquid chromatography(HPLC)with ultraviolet detection.The effects of processing on 18 components such as aspartic acid in Platycladus orientalis leaves were studied,which provided a reference basis for further quality evaluation standards of Platycladus orientalis leaves.4?The determination method of quercetin in POL was established by ultraviolet colorimetry,and the content of total flavonoids in POL was compared by different extraction methods.According to this method,the most suitable extraction process of flavonoids in POL was studied.5?To observe the antipyretic,anti-inflammatory,analgesic and bacteriostatic effects of flavonoids in the leaves of Platycladus orientalis.Material and method:1?The liquid phase column is Yuexiu extreme(long-acting carbon 18,5 micron,250mm).The sample eluent is methanol-water(0.2%phosphoric acid)gradient elution.The UV detection wavelength is 330 nm,and the sample eluent velocity is 1 mL/min.2?According to the pharmacopoeia(general rule 0213)according to carbon method,according to speculation method processing.3?HPLC was used for the determination of 18 kinds of amino acids in the samples.Acetonitrile-water(1:1):4%sodium acetate buffer salt was used as the mobile phase,the flow rate was 1.2mL/min,the gradient elution,the detection wavelength was 360nm,and the column temperature was 27?.4?Ultraviolet spectrophotometry was used to determine the absorbance at 423 nm with quercetin as index and aluminium chloride colorimetric method.Four processes were used:degreasing with petroleum ether and extracting with 75%ethanol,extracting directly with 75%ethanol,extracting with 50%ethanol after water extraction and ethanol precipitation,and 70%ethanol after water extraction and ethanol precipitation.5?The extraction rate of total flavonoids from Platycladus orientalis leaves was studied by orthogonal test with four factors and three levels.The effects of extraction solvent,extraction times,extraction time and solid-liquid ratio on the extraction rate of total flavonoids from Platycladus orientalis leaves were studied.To determine the best extraction process of Platycladus orientalis leaves.6?The total flavonoids of Platycladus orientalis leaves were extracted by the established preparation process,and the antipyretic effect was studied by 2,4 dinitrophenol-induced fever in rats;the anti-inflammatory effect was observed by egg white and xylene-induced inflammation;the analgesic effect was observed by hot plate method in mice;and the bacteriostatic effect was observed by bacteriostasis in vitro.Results:1?The correlation coefficients of standard curves of myricetin,kaempferol,quercetin,myricetrin,amentoflavone,hinokiflavone and quercitrin were more than or equal to 0.9995,indicating that the linear relationship was good,and the average recovery rate and relative standard deviation met the relevant requirements(n=6).2?After processing,the contents of myricetrin,quercitrin,hinokiflavone and amentoflavone decreased,while the contents of myricetin,quercetin and kaempferol increased.3?The medicine herbs included all of the detection of 18 amino acids,included 8 kinds of essential amino acids from different origins.The contents of the highest essential amino acids was in Hebei and total amino acids was in Anhui.18 amino acids were decreased after processing,and with the degree of deepening the amino acid contents decreased significantly.4?the content of flavonoids in the extract was 70%.5?The optimum extraction conditions were microwave time 90s,methanol concentration of 70%and solid-liquid ratio of 1:40.6?Flavonoids in Platycladus orientalis leaves inhibited fever induced by 2,4-dinitrophenol in rats,inflammation induced by xylene and egg white,and pain induced by heat stimulation.In vitro bacteriostasis experiments showed that they had different inhibitory effects on Staphylococcus aureus and Dicoccus pneumoniae.Conclusion:1?The method is simple,precision,recovery,repeatability can meet the requirements,and can be used for determination of the content of the Cacumen Platycladi and Cacumen Platycladi Carbonisatum2?The method is simple,can be used for determination of the content of the Cacumen Platycladi and processed Cacumen Platycladi.The results of the content shows that the flavone in Cacumen Platycladi changed by processing.3?The contents of amino acids in the medicinal materials of platycladus orientalis leaves has a certain significance to evaluate its quality.Prepared the contents of amino acids before and after being processed the difference was obvious,can be considered without processing directly into the medicine.4?Conclusion:the aluminum chloride colorimetric method was stable and reliable,accurate determination of total flavonoids in leaves of Platycladus orientalis.5?The present investigation provided the optimum extraction process of the total flavonoids and quercitrin from the Platycladus orientalis Franco.6?Flavonoids from Platycladus orientalis leaves have good antipyretic,anti-inflammatory,analgesic and bacteriostatic effects.
