Purpose:Through collections of 5 classical prescription which contains radix ophiopogonis of modern clinical effective cases,using modern statistical methods,combining with the radix ophiopogonis efficacy of materia medica literature research,exploring the 5 classical prescription radix ophiopogonis clinical application of commonly used dose,efficacy function,the relationship between the effects of disease and compatibility mechanism associated with the core of the formulas.It provides theoretical basis and data support for the clinical application of radix ophiopogonis in 5 classical prescription,and provides new ideas and methods for the clinical dose-effect relationship research of TCM.Methods:1.Through CNKI,searched the medical cases with the name of five classical prescription as keywords between the establishment of the journal and January 1,2020,sorted out the effective cases of the modern application of classical prescription in the treatment of diseases,and summarized the historical history of the efficacy of radix ophiopogonis and the related disease syndromes and doses.2.Input the collected medical case information into Excel and build a database.3.SPSS 22 frequency analysis was used to calculate the high-frequency main diseases,syndromes,main symptoms,concurrent symptoms,tongue and pulse,drug taste,drug frequency and drug dose of the five prescriptions.T test was used to compare the dosage of radix ophiopogonis with that of radix ophiopogonis in han dynasty.Correlation analysis was used to study the correlation between the amount of ophiopogon japonicus and the total prescription dose and the total prescription taste.Variance ANOVA was used to analyze the difference in the amount of ophiopogon in the five prescriptions,the difference in the amount of ophiopogon as a royal medicine and a non-royal medicine,and the difference in the ratio of the amount of ophiopogon in the total amount of the prescriptions.4.SPSS Modeler 14 Apriori model system was used to carry out correlation analysis on the high-frequency drugs,symptoms,syndromes and ophiopogonum dosage of the five prescriptions respectively,and the strong effective correlation drug combination containing ophiopogonum and the dose-effect correlation law of disease and syndrome were obtained.Results:1.Through searching on the Internet,a total of 566 cases of qualified modern medicine prescriptions were collected,including 107 cases of maimendong decoction,113 cases of zhuyeshigao decoction,221 cases of zhigancao decoction,94 cases of wenjing decoction,and 31 cases of shuyu decoction.After frequency analysis of effective medical case data,it is concluded that:The main high-frequency diseases of maimendong decoction were cough,plum kernel qi,asthma,lung dysfunction,epistaxis.The high frequency syndrome is Yin deficiency syndrome,Yin deficiency fire flourishing syndrome,qi Yin deficiency syndrome.Typical prescriptions are cough,vomiting,epigastric pain,dry mouth,expectoration of phlegm,poor tolerance,less red tongue moss,and pulse size.The number of prescriptions was concentrated between 7-1 1,and the highest frequency was 8.The main high-frequency diseases of zhuyeshigao decoction were fever,oral sores,insomnia,vomiting and hiccup.The high frequency syndromes were qi and Yin deficiency syndrome,stomach heat syndrome,Yin deficiency and fire flourishing syndrome.Typical prescriptions were thirst,god fatigue,upset,tongue red moss less,pulse number;The number of prescriptions concentrated in 5-7,the highest frequency was 5.The main high-frequency diseases of zhigancao decoction were palpitations,premature ventricular beats,viral myocarditis and insomnia.The high frequency syndromes are qi and Yin deficiency syndrome,qi and blood deficiency syndrome,Yin and Yang deficiency syndrome.The typical prescriptions were palpitation,chest tightness,fatigue,shortness of breath,dizziness,pale tongue coating,thin white,and pulse generation.The number of prescriptions was distributed between 10 and 12,and the highest frequency was 10.The main high-frequency diseases of wenjing decoction were dysmenorrhea,infertility,various syndromes before and after dysmenorrhea,amenorrhea;The high frequency syndromes were blood cold syndrome,blood stasis syndrome and deficiency of qi and blood.The typical prescriptions were abdominal pain,no pregnancy after marriage,less menstruation,chills,thin white tongue moss,and fine pulse.The number of prescriptions was mainly 12.The main high frequency diseases of shuyu decoction are cancer,asthenia,vertigo;The high frequency syndrome is deficiency of qi and blood,deficiency of Yin and Yang.Typical prescription syndrome is fatigue,thin,dizzy,lumbar acid,insomnia,pale tongue coating thin white,pulse heavy fine;The number of prescriptions was concentrated at 21.2.The main effects of ophiopogonum in the five first formula probably are:Clearing heat and benefiting pharynx(maimendong decoction),Nourish Yin and relieve vomiting(zhuyeshigao decoction),regulating blood circulation and dispersing blood stasis(zhigancao decoction),nourish Yin and moisten dryness(wenjing decoction),invigorate qi and strengthen Yin to replenish deficiency(shuyu decoction).3.The average amount and the most common amount of ophiopogon in the five prescriptions were maimendong decoction 25.Og and 30g respectively.Zhuyeshigao decoction 16.1g and 15g;zhigancao decoction 15.1g and 15g;wenjing decoction 15.3g and 10g;shuyu decoction 12.6g and 10g.The mean dosage of ophiopogon was used for analysis of variance.Compared with other four groups(ophiopogon is not sovereign drug),the difference was statistically significant(P<0.01).The comparison between the zhuyeshigao decoction group,the zhigancao decoction group and the wenjing decoction group showed no statistical difference(P>,0.05).Compared with the other four groups,P<0.05 was statistically significant.The mean specific weight of ophiopogon japonicus in the whole recipe was 20.19%(maimendong decoction),12.26%(zhuyeshigao decoction),10.33%(wenjing tang),8.32%(zhigancao decoction)and 5.46%(shuyu decoction).The results of variance analysis were:F value=96.46,P<0.05,the proportion of ophiopogon japonicus was different among the parties.The t-test analysis of the modern dosage of radix ophiopogonis in the fourth and fifth prescriptions and the dosage in the han dynasty showed P<0.05,indicating that there was a significant difference in the ancient and modern dosage.The correlation analysis showed that the amount of ophiopogon japonicus was correlated with the total prescription dose,P<0.05.No significant correlation was found with the number of prescriptions,P>0.05.4.Through correlation analysis of high-frequency drugs in each group,24 groups of related drug combinations including ophiopodium were selected according to the support degree>20%,confidence degree>80%,and promotion degree>1.The two drug pairs of wheat mendong decoction include:ophiopogone-chinese yam;Ophiopogon japonicus;Ophiopogon-codonopsis pilosula.The three drug groups included:ophiopogon japonicus-Chinese yam and pinellia japonicus;Ophiopogon japonicus and pinellia japonicus;Radix ophiopogonis-japonica rice and radix codonopsis codonopsis.Bamboo leaf gypsum decoction two drug pairs include:ophiopogonis-japonica rice;Ophiopogon-codonopsis pilosula.The triple drug group included:ophiopogon japonica rice and gypsum;Ophiopogon japonica rice and pinellia japonica;Ophiopogon-codonopsis and bamboo leaves.Cong-glycyrrhiza decoction two drug pairs include:ophiopogonis-cassia twig.The three drug groups include:radix ophiopogonis-cassia twig and processed licorice root;Radix ophiopogonis-cassia twig and radix rehmanniae;Roasted licorice-dates and ophiopogon.Wen jing soup two medicine pairs include:ophiopogonum-pinellia.The three drug groups included:ophiopogon japonicus-pinellia and angelica sinensis;Ophiopogon--pinellia and evodia;Ophiopogonum-pinellia and cortex moutan.Dioscorea dioscorea pill two drug pairs include:ophiopogonum-cassia twig;Ophiopogon-codonopsis pilosula.The three drug groups included:radix ophiopogonis-cassia twig and angelica sinensis;Ophiopogon--radix codonopsis and Chinese yam;Radix ophiopogonis-cassia twig and radix bupleuri.5.According to>5%support degree,>50%confidence degree and>1 promotion degree,20 effective and strong correlation laws were obtained by correlation analysis of ophiopods dosage in each differences in the amount of ophiopogon japonicus in maimendong decoction,zhuyeshigao decoction,zhigancao decoction,wen jing decoction and shuyu decoction.There was a significant difference in the dosage of ophiopogon japonicus as a sovereign drug and a non-sovereign drug.There is a significant difference between the dose of classical prescription ophiopogonis in han dynasty and the dose of jingfang ophiopogonis in modern application.The dosage of ophiopogon japonicus was correlated with the total dosage.The effect of ophiopogonum in 5 classical prescription was affected by the compatibility environment and dosage.The efficacy and dosage of radix ophiopogonis in classical prescription are probably as follows:the dosage of radix ophiopogonis above 25g(large dose)plays the role of clearing heat,relieving pharynx and relieving cough,15-25g(medium dose)plays the role of reducing adverse pulse and blood circulation,and dosage under 15g(small dose)plays the role of nourishing Yin,nourishing qi and filling deficiency.
