Purpose By observing the effects of Gubitong Prescription on cartilage histopathology and Wnt5a,?-catenin,MMP-3,Bax gene and protein expression in osteoarthritis rats,we explore Gubitong Prescription's regulation of Wnt/?-catenin signaling pathway to interfere osteoarthritis's mechanism of chondrocyte apoptosis.Method Six-week-old SD rats were selected and randomly divided into normal group,sham operation group,model group,positive drug control group,low-dose group of Gubitong Prescription,middle-dose group of Gubitong Prescription and High-dose group of Gubitong Prescription.The rats in the normal group did not intervene;the sham operation group only cut the skin around the joints and exposed the joints;the model group,the positive drug control group,the low-dose group of Gubitong Prescription,the medium-dose group of Gubitong Prescription and the high-dose group of Gubitong Prescription was modeled by Hulth method.After successful modeling,the normal group,sham operation group and model group were given saline,the positive drug control group was given glucosamine hydrochloride,Gubitong Prescription low-dose group,Gubitong Prescription medium-dose group and Gubitong Prescription high-dose group was given Gubitong Prescription low,medium and high-dose gavage,and each group was gavaged continuously for 6 weeks.The SD rats were sacrificed,and the left(surgical side)knee joints were removed for HE staining,q-PCR and Western blot detection.The HE staining ManKin's score results and Wnt5a,?-catenin,MMP-3,Bax genes and proteins' expression were observed between each group.Result Comparing the ManKin's scores of HE staining in each group,it was found that there was no significant difference between the ManKin 's scores of the normal group and the sham operation group(P>0.05).The ManKin's score of the articular cartilage of the model group was higher than that of the sham operation group,and the difference was statistically significant(P<0.05).Compared with the sham-operated group,the positive drug control group,Gubitong Prescription low-dose group,Gubitong Prescription medium-dose group and Gubitong Prescription high-dose group were all increased,and the differences were statistically significant(P<0.05).Compared with the low-dose group of Gubitong Prescription,the medium-dose group of Gubitong Prescription,and the high-dose group of Gubitong Prescription,the positive drug control group had a significantly higher ManKin's score(P<0.05).Compared with the low-dose group of Gubitong Prescription,the medium-dose group of Gubitong Prescription and the high-dose group of Gubitong Prescription,there was no statistically significant difference in ManKin 's score between the groups(P>0.05).The results of q-PCR and Western blot showed that there was no significant difference in the relative expression of Wnt5a,?-catenin,MMP-3,Bax genes and proteins between the normal group and the model group(P>0.05).Compared with the sham operation group,the relative expression levels of Wnt5a,?-catenin,MMP-3,Bax genes and proteins were increased in the model group,and the difference was statistically significant(P<0.05).Compared with the sham operation group,the relative expression levels of Wnt5a,?-catenin,MMP-3,Bax genes and proteins increased in the positive drug group,and the difference was statistically significant(P<0.05).Compared with the model group,the relative expression levels of Wnt5a,?-catenin,MMP-3,Bax genes and proteins were reduced in the sham operation group,and the difference was statistically significant(P<0.05).Compared with the low-dose group of Gubitong Prescription,the medium-dose group of Gubitong Prescription and the high-dose group of Gubitong Prescription,the relative expression of Wnt5a,?-catenin,MMP-3,Bax genes and proteins increased in the positive drug group The difference was statistically significant(P>0.05).Compared with the low-dose group of Gubitong Prescription,the medium-dose group of Gubitong Prescription and the high-dose group of Gubitong Prescription,the relative expression of Wnt5a,?-catenin,MMP-3,Bax genes and proteins was not statistically significant(P>0.05).Conclusion Gubitong Prescription can inhibit the activation of Wnt/?-catenin signaling pathway,down-regulate the expression of Wnt5a,?-catenin,MMP-3 and Bax,play a role in protecting articular cartilage and inhibiting chondrocyte apoptosis.
