Objective:This article intends to explore the MGF secretion in rat skeletal muscle and heart function improvement effect on mechanical vibration or intermittent aerobic exercise to promote MI,provide a scientific basis for the exploration of safer and more effective rehabilitation programs for the effective treatment of myocardial infarction.To exercise improves ischemic heart disease provide a new theoretical basis.Materials and Methods:3-month-old male SD rats were 48,respectively,sham surgery and myocardial infarction surgery,were randomly divided into four groups,each group 12:Quiet sham MI group(Sham group),myocardial infarction quiet group(MI group),myocardial infarction + high-intensity interval aerobic exercise group(ME group),myocardial infarction + intermittent mechanical vibration group(MV group).Established rat model of myocardial infarction using the left anterior descending coronary artery ligation.Rat intermittent aerobic exercise treadmill:ME group 1wk convalesce after surgery began treadmill training,adaptive training the first week to 10 m/min velocity 30min;Second week of formal training to speed 10m/min warm 10 min,then to velocity 25m/min 7min(exercise intensity 85%-90%V02max),and 15 m/min speed of movement 3min(exercise intensity 50%-60%V02max),sequentially alternating,the total time of 60min,weekly training 5d,training lasted 8 wks.Using self-developed mechanical shaker small animals vibration training:rats in the first week after MV adaptive vibration training to 15Hz,15min,1t/d,5d/wk;second week,officially vibration training,frequency;25Hz,amplitude:2mm,time:15min,2t/d,the middle two intermittent 8min,the total time of vibration training for 38min,5days/week,lasted 8wks.Using PowerLab 8/30 bio-signal analysis system to measure hemodynamics,heart rate and ECG indicators;Using paraffin sections,Masson,TTC and HE staining and morphological parameters measure the heart and skeletal muscles;Using immunohistochemistry and Western Blot method of observation and determination of a-actin protein expression in the myocardium and a-myosin;Measured by ELISA MGF gastrocnemius and serum protein;Determination of the gastrocnemius muscle and cardiac muscle MGF using Western Blot method,ERK1/2,p-ERK1/2 and MEK1/2 expression;Gastrocnemius muscle was measured using RT-qPCR myocardial MGF mRNA expression;Using SPSS 17.0 for Windows software package for Pearson correlation analysis.The results showed that:(1)Mechanical vibration or intermittent aerobic exercise can effectively reduce infarct size,reducing MI cardiac collagen deposition.Intermittent aerobic exercise or mechanical vibration intervention can significantly reduce infarct size was significantly reduced infarct zone collagens,mechanical vibration intervention effect is more pronounced;(2)Mechanical vibration or intermittent aerobic exercise can improve heart function in MI rats,and the mechanical vibration safer.After intermittent aerobic exercise intervention was 67%survival rate of rats;Survival after mechanical vibration intervention was 92%,indicating that the load on the heart of mechanical vibration training produced little of myocardial infarction in rats safer.Two interventions could significantly improve cardiac function in rats MI,and mechanical vibration safer;(3)Mechanical vibration or intermittent aerobic exercise significantly improved the survival of rat cardiac contractility MI protein levels.MI group significantly reduced myocardial infarction fringe a-actin protein expression and a-myosin;increased significantly after the mechanical vibration or intermittent aerobic exercise intervention,mechanical vibration effect is more significant;(4)Mechanical vibration or intermittent aerobic exercise can improve MI rat gastrocnemius muscle cross-sectional area,relieve myocardial infarction-induced muscle atrophy and intermittent aerobic exercise is superior to mechanical vibration effects;(5)Mechanical vibration or intermittent aerobic exercise can improve rat gastrocnemius MI MGF expression and serum levels,and promote skeletal muscle synthesis and release into the blood,the corresponding target organs play a biological role of positive significance;(6)Mechanical vibration or intermittent aerobic pathway activation gastrocnemius MGF/MEK1/2/ERK1/2 play the corresponding biological function,relieve myocardial infarction-induced muscle atrophy and intermittent aerobic exercise is superior to mechanical vibration effects;(7)Mechanical vibration or intermittent aerobic exercise significantly increased the expression of MI myocardial MGF,and intermittent aerobic exercise is superior to mechanical vibration effects;(8)Mechanical vibration or intermittent aerobic exercise myocardial MGF/MEK1/2/ERK1/2 activation pathway exerts its biological function,causing cardiac hypertrophy corresponding myocardial infarction survivors and mechanical vibrations better than intermittent aerobic exercise effect;(9)Mechanical vibration or intermittent aerobic exercises stimulate the MI rat gastrocnemius MGF upregulation and release into the blood;(10)Mechanical vibration or intermittent aerobic stimulation of rat gastrocnemius and upregulation of serum MGF MI and cardiac pERK1/2 expression are highly correlated;(11)Mechanical vibration or intermittent aerobic stimulation of rat gastrocnemius and upregulation of serum MGF MI lifting height associated with cardiac function.Conclusion:1 Mechanical vibration or intermittent aerobic exercise can effectively reduce infarct size,significantly improved the survival of myocardial contractile proteins in rat MI levels,improve heart function in MI rats,while the mechanical vibration safer and more effective;2 Mechanical vibration or intermittent aerobic exercise can improve MI rat gastrocnemius muscle cross-sectional area,relieve myocardial infarction-induced muscle atrophy and intermittent aerobic exercise is superior to mechanical vibration effect.May be associated with skeletal muscle MGF/MEK1/2/ERK1/2 pathway activation;3 Mechanical vibration or intermittent aerobic exercise significantly increased the expression of MI myocardial MGF,effectively improve the heart function,may increase by stimulating the secretion of rat skeletal muscle MGF MI,improve blood concentrations of MGF,circulation to distant organs heart,great protection MI rat heart function,or directly activate cardiac MGF/MEK1/2/ERK1/2 pathway exerts its biological function,causing cardiac hypertrophy corresponding myocardial infarction survivors and mechanical vibrations better than intermittent aerobic exercise effect;4 Mechanical vibration or intermittent aerobic exercises stimulate the MI rat gastrocnemius MGF upregulation,lifting height associated with myocardial ERK1/2 phosphorylation and cardiac function.
