We have designed and synthesized a completely novel carbohydrate based cancer vaccine candidate by replacing carrier proteins with a zwitterionic polysaccharide, PS A1 1. The mouse studies and in vitro human cancer cell studies by flow cytometry against our vaccine showed T-cell dependence and specific antibody production against tumor associated Tn hapten. These results also show that a chemical modification of zwitterionic polysaccharide PS A1 1 does not alter its immune response. Our vaccine has opened a completely new area in the field of glycoimmunology. We have also developed a one-pot, two-step reaction protocol for the synthesis of regiochemically differentiated 1,2,4,5-tetrahydro-1,4-benzodiazepin-3-ones. These biologically relevant small molecules can be prepared efficiently with the help of microwaves under environmentally benign reaction conditions. |
