Azo dye wastewater has high chroma and complicated component,which always poses a serious threat to environment for producing highly carcinogenic aromatic amines organics in degradation processing.Biological treatment for Azo dye removal from wastewater is a kind of high efficiency and low cost methods.In this work,one fungus,with high efficiency of degradation to azo dyes-mordant yellow 1,was screened from activated sludge.Degradation of simulative dyeing wastewater and the best immobilization method and conditions were optimized.Results are as follows:(1)One fungus TS-A was screened from activated sludge,Through morphological,physiological and 18S r DNA identification,the fungus was identified and named Aspergillus TS-A.Resulted showed that decolorization rate was 83%from simulated wastewater.(2)The paper preliminary study the degradation characteristics of TS-A degreasing mordant yellow 1.Experiments to determine the best degradation conditions of TS-A as follows:p H 6.0;temperature 35°C;carbon-to-nitrogen ratio,30:4,Under optimized conditions,the decolorization rate was 89%.Results showed that metals including Ba2+?Sr2+?Ca2+?Zn2+?Co2+?Cs+,employed a promoting effect on the TS-A decolorization of mordant yellow 1.Moreover,Fe3+?Al3+?Mg2+?Cu2+played a promoting role on microbial decolorization in 24h,with the extension of time,the metal ions accumulated in microorganisms and metal ions on microbial decolorization played a role in inhibition.On the contrary K+?Na+?Mo4+?Ni2+?Mn2+?EDTA showed obvious inhibition on microbial decolorization.(3)Fourier transform infrared chromatography(FTIR)and Gas Chromatography-Mass Spectrometer(GC-MS)were adopted to analysis chemical composition of the fermentation broth and speculated the way of TS-A degradating mordant yellow 1.Rresults showed that TS-A could not only break out luminescent groups azo group(-N=N-),but also have a high recognition rate for C-N.what's more,TS-A degradation of azo dyes could not produce aromatic amines.(4)TS-A was immobilized with method of adsorption,and immobilized carrier was selected with decoloration rate as the index.Results showed that the immobilized TS-A decolorization of mordant yellow 1 had the best effect when macroporous polyurethane foam acted as the carrier.Single-factor method was applied to screen the condition of immobilized TS-A decolorization.The best optimum conditions of immobilized TS-A as follows:p H 6.0;temperature 35°C;carbon-to-nitrogen ratio,30:3.Metal ions strengthing immobilized TS-A decolorization of mordant yellow 1 wastewater rate could reach 100%under the best optimum conditions.The results showed that,improving degradation rate of TS-A though optimization decoloring conditions and immobilization technology.These make it possible to ues TS-A as the main mordant yellow 1 degradation microbial to manage industrial environment.
