Endothelial cells(ECs)are the natural barrier between blood and blood vessel walls,with significant apico-basal polarity.When different stimulating factors(IL-1?,LPS,PMA)stimulate endothelial cells on both sides of the monolayer,LPS and PMA produce a significant polarity response.However,PMA(Phorbol-12-myristate-13-acetate)as a hydrophobic small molecule polar response phenomenon has aroused our curiosity.PMA is frequently used as a PKC activator in biological studies because it can replace PKC natural activator DAG in vivo.PKC plays an important role in the maintenance of tight junction protein phosphorylation of endothelial cell polarity.We confirmed that the polarity response of endothelial cells may be related to the polarity distribution of PKC by the detection of intracellular protein(MCP-1,IL-8,IL-6),secretion and confocal imaging.By detecting the expression and release of inflammatory cytokines and analyzing the relationship between them,it`s shown that PKC-mediated cell polarity response is related to inhibition of basal cytokine release.Further studies have found that PKC ? and PKC ? subtypes play a major role in the process of polar secretion of inflammatory factors.However,PKC specific mechanism for the regulation of this process remains to be further studied.
