JMJD5,also known as KDM8,is a member of the histone demethylase family(also called KDM family)containing the JmjC domain.It has been reported that JMJD5 is a microtubule-associated protein in mouse mitosis,and it is essential for the normal mitosis progression.The biological function of JMJD5 in mouse meiosis remains unknown.In this experiment,we examined the expression,subcellular localization and function of JMJD5 in mouse oocytes meiosis.The results of this experiment indicated that JMJD5 was stably expressed during the key periods of meiosis.JMJD5 was located in the germinal vesicle at the germinal vesicle stage,and as the germinal vesicle burst,JMJD5 was located around the chromosome.In addition,JMJD5 was located on the entire spindle at pro-metaphase I stage to metaphase II stages.The interference treatment of nocodazole on oocytes further demonstrated that JMJD5 was co-localization with ?-tubulin.To further explore the relationship between JMJD5 and microtubule,and find the mechanism of actin,the specific JMJD5 short interfering(si)RNA was injected into mouse oocytes to deplete JMJD5 expression.The results showed that depletion of JMJD5 expression caused abnormal spindle assembly and migration,misaligned chromosomes,blocked cell cycle,and reduced filament assembly.At the same time,the normal localization of ?-tubulin was severely disrupted and the spindle checkpoint protein BubR1 was activated.Therefore,this experiment proves that JMJD5 is a member of the microtubule organizing center,which regulates the spindle assembly,establishes the cortical area polarity,and switches at the metaphase I-anaphase I stages.
