The dopamine system is an indispensable neuroregulatory center in the animal kingdom(including humans).Its secretion is closely related to various social behaviors such as human lust and feelings,and it conveys the message of excitement and happiness.At present,5-HT neurotransmitter system has been widely studied in the study of the neural structure formed by sexual preference,and several literatures have confirmed that the decrease of 5-HT level in the mouse brain will lead to the change of male sexual preference.However,the relationship between the dopamine system,which is directly involved in reproductive and social behavior in species,and the formation of sexual preference has been rarely reported.At present,I have started from the dopamine system,used chemical genetics,optogenetics and other means to conduct reverse virus tracking experiment,and then established the relationship between the testis and the dopamine system;G-camp calcium imaging experiment was conducted to determine the activity of dopamine neurons in mice before and after exposure and during exposure.At the same time,microdialysis experiments were carried out in the brain area of VTA in awake individuals to quantitatively observe the dopamine activity during the behavioral behavior of mice.Combined with genetic manipulation experiments,electrophysiology,electrochemistry,ultrasonic monitoring and confocal,the specific mechanism of how the dopamine system determines the formation of bias was studied.Here we have pioneered the central mechanism by which the dopamine system is responsible for determining preference formation.1.In vivo,by changing the dopamine level of mouse VTA(Ventral tegmental area),the preference of male mice for female can be reduced,and a large amount of time is transferred to sniff male mice,accompanied by typical back-crawling behavior.2.The C57BL/6J mice were implanted in the VTA area and injected with the dopamine receptor D2 antagonist,and the preference of the mice for invasive males was significantly increased.3.Based on the laboratory study of Syt11,it was established that Syt11 could affect the upper membrane of dopamine receptor D2 and increase the overall level of dopamine.Therefore,through the construction of Syt11 c KO mice,we found that the male mice had decreased fertility,decreased mating strength and changed sexual preference to female mice in behavioral observation.4.Through the optogenetic and chemo genetic manipulation of Syt11 c KO mice,we can initiate the recovery of female sexual preference by inhibiting VTA dopamine levels.
