The disease tissue research model is not only an important supplement to the current preclinical research model,but also a hot and difficult point in the field of biomedical research.The development of 3D culture technology makes it possible to simulate living tissues and organs in vitro,so that disease research is no longer limited to traditional research models,and promotes the common deve lopment of disease pathology,cell biology,precision medicine,drug toxicity and efficacy detection and other fields.In this study,3D in vitro disease tissue research models were prepared using 3D culture technology which including tumor tissue and adipose tissue.Normal stromal cells were activated to express tumor-related microenvironment characteristics through in vitro conditional culture,tend to provide a microenvironment basis for the establishment of 3D tumor tissues.Through m RNA sequencing and drug testing,we confirmed that 3D tumor tissues not only have high similarity with tumor tissues in vivo in terms of metabolic and malignant transformation characteristics,but also have potential for drug screening and can serve as an organizational plat form for high-throughput drug screening.In addition,an in vitro adipose tissue based on bacterial cellulose scaffold was established,which can be used not only as a paracancerous tissue for tumor research,but also as a research model for metabolic-related diseases.Primary culture using living tissues and rapid expansion on bacterial cellulose showed similar in vivo lipid differentiation behavior.By in vivo implantation,the 3D adipose tissue can retain the basic cell type of adipose tissue for a long time and have good chimerism with host tissue.The tissue model established in this study has good performance in biomimetic simulation of histological characteristics and physiological functions,which can provide an in vitro research tool for tumor and f at-related pathological studies,and lay a foundation for the establishment of a clinical tissue sample biobank.
