The cellular environment is complex,the use of small-molecule fluorescent probes has not only achieved the cellular-level imaging,but also achieved subcellular imaging,enabling researchers get in the biological studies to a further level.As the specific application changes,the requirements for small-molecule fluorescent probes are higher so that they are constantly optimized.The fluorescent response-probes which under a certain condition are more sensitive and selective.These responsive probes are more intelligent and reduce the background fluorescence.In the case of many kinds of biochemical reactions in cells,the bio-orthogonal reactions can also react with rapidly,selectively and high yield.Among these bio-orthogonal reactions,photoclick reaction which diaryltetrazole react with an alkene dipolarphile under photoirradiation could afford the pyrazoline cycloadducts.The characteristics of photoclick reaction are rapid rate,using light instead of metal ions as catalyst to reduce cell toxicity,in addition to this the reactant diarytetrazole is non-fluorescent,the fluorescent of pyrazoline "turn on" under the photoirradiation.1.All diaryltetrazole compounds are synthesis with the arene diazonium salts and sulfohydrazones.Compound la-1g are intended to react with nucleoside analogue,5-vinyl-2'-deoxyuridine(VdU)for nuclear imaging.And the compound Tet? and Tet? which could carry out intramolecular photoclick reaction modified with organelle anchoring motifs for mitochondrial or lysosomal imaging.2.The chemical structure of all compounds were identified by NMR and ESI-MS.The spectral properties and cytotoxicity were confirmed.Compounds la-1g and VdU have poor reactivity in aqueous solution.As for the intramolecular photo click reaction of Tet ? and Tet ? are more reactive in aqueous solution and afford fluorescent product just with a hand-held UV lamp in living cells.The imaging of cellular mitochondria and lysosome is achieved with high colocalization coefficients.In summary,it determined that reactivity between compound la-1g and VdU and the intramolecular photo click reaction of Tet ? and Tet ?.Tet ? and Tet ? could be lighted up with hand-held UV lamp to achieve mitochondrial or lysosomal imaging.
