Influenza is a large-scale epidemic disease that is seasonal and highly contagious and seriously endangers the safety of human society.It is very important for the prevention and control of influenza,especially avian influenza.Avian influenza virus,as a pathogen that causes humans to suffer from highly pathogenic avian influenza,will cause fever,diarrhea,muscle aches and other symptoms similar to ordinary influenza after infecting the human body.Unlike the seasonal flu caused by the usual human influenza virus,the avian influenza virus gains cross-species infectivity after genetic recombination,and the human body has no initial immunity to such virus subtypes,so when the human body is first exposed to the avian influenza virus After that,it will trigger a more serious body immune response than ordinary flu,and in severe cases,it may even cause damage to the body.Therefore,planned prevention of the avian influenza virus will have a better effect than post-infection treatment.As an RNA virus,avian influenza virus has a high mutation rate and highfrequency recombination characteristics in its genome.At present,most influenza vaccines use one of the virus' s characteristic proteins: hemagglutinin HA protein to expose the head of the antigen design.This led to the difference between the influenza vaccine strain and the virus epidemic strain in the current season.At the same time,the broad spectrum of vaccines is also insufficient.The latest research found that the amino acid sequence of the stem region of the protein intersects the head region of the HA protein and is relatively conserved.No obvious mutations were found in viruses of multiple homologous subtypes.In addition,most of the previous influenza vaccines were inactivated and attenuated vaccines.The adjuvants and other compounds used therein had problems such as unknown side effects,and their stability and safety could not be guaranteed.Currently,they can only be widely used in poultry.In this project,we will use the epitope vaccine,one of the latest vaccine technologies,to design and screen suitable HA protein epitope sequences for H9N2 avian influenza virus.The use of highly conserved region-specific antigens predicted by bioinformatics in place of the antigens in the highly variable regions of the head can improve the cross-protection ability of avian influenza vaccines;animal experiments have verified the effectiveness of the designed antigen peptides;Subsequent recombinant virus-like particles provide antigen information of the H9N2 avian influenza virus with high antigenicity.In addition,this topic will also test the practicability of the designed antigen screening method,and provide an effective method for subsequent antigen screening of other subtypes of avian influenza viruses.
