In recent years,food poisoning caused by Vibrio parahaemolyticus has become more and more serious,which seriously affects human health.Therefore,the development of natural and effective food antibacterial agents has become important.As a natural food additive,sulfur-containing flavor is not only a common flavoring substance in food,but also has antibacterial effects,and has attracted people's attention.In this paper,benzyl isothiocyanate(BITC)was selected as the research object.The differentially expressed genes of V.parahaemolyticus were screened by transcriptome sequencing,and the Gene Ontology(GO)classification and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed.The real-time quantitative PCR(q RT-PCR)method was used to determine virulence gene expression to verify the accuracy of transcriptome data.Meanwhile,changes in swimming ability,swarming ability,biofilm formation,and cell membrane permeability were examined.The results showed that under the treatment of BITC with a sub-inhibitory concentration of 1/4 MIC,there were 332 differentially expressed genes,of which 195 were up-regulated and 137 were down-regulated.507 differentially expressed genes were obtained under the 1/8MIC BITC treatment,of which 234 were up-regulated and 273 were down-regulated.The GO enrichment analysis found that under the treatment of 1/4 MIC BITC,the genes were annotated into 1189 GO terms,of which 641 were annotated into biological processes,168 were in cellular components,and 380 were in molecular function.Under the treatment of 1/8MIC BITC,genes were annotated into 1679 GO terms,of which 891 were annotated into biological processes,227 were annotated into cellular components,and 561 were annotated to molecular function.Through the analysis of KEGG pathway of differential expressed genes,it was found that BITC treatment plays an important role in metabolic pathways,biosynthesis of secondary metabolites,microbial metabolism in different environments,and carbon metabolism pathways.At the same time,BITC can significantly inhibit the virulence gene expression of Vibrio parahaemolyticus.The results of q RT-PCR are as same as the transcriptome results.BITC can effectively inhibit its swimming ability,inhibit biofilm formation,and change the permeability of cell membrane.In summary,BITC has a good antibacterial effect on Vibrio parahaemolyticus,which can be used as a potential natural food additive in the food industry,prolonging the shelf life of food,and has good control and prevention against foodborne diseases.
