China is one of the countries with the most serious drought in the world.Zygophyllum xanthoxylon is a super xerophytic plant which grows in desert and gobi regions and has strong drought tolerance.The OSCA,NAC,bZIP and WRKY gene families play a role in drought resistance in plants,and the identification and analysis of these families are focused on model species.Virus-induced gene silencing(VIGS)can verify the function of genes,and it mainly constructs the VIGS system in model species,while it is seldom studied in Zygophyllum xanthoxylon.So we analyzed the four gene families in Zygophyllum xanthoxylon.Based on this study Zygophyllum xanthoxylon transcriptome data,some bioinformatics methods were used to analyze the classification,physicochemical properties,secondary structure,conserved motif and phosphorylation sites of OSCA,NAC,bZIP and WRKY families members.The transmembrane domain of ZxOSCA gene family was analyzed,ZxOSCA1.3 gene function verification,construction VIGS vector,grope for Zygophyllum xanthoxylon VIGS system.The main results of this study are as follows:1.9 OSCA genes,28 NAC transcription factors,22 bZIP transcription factors and 45 WRKY transcription factors were identified in the Zygophyllum xanthoxylon transcriptomic data,and the protein sequences of these genes were sorted through the evolutionary tree.ZxOSCAs was divided into 4 subfamilies,ZxNACs into 8 subfamilies,ZxbZIPs into 9 subfamilies,and ZxWRKYs into 6 subfamilies.2.The physical and chemical properties of ZxOSCAs sequence showed little difference,and the proteins were alkaline and hydrophobic.The physical and chemical properties of ZxNACs,ZxbZIPs and ZxWRKYs,such as the number of amino acids and isoelectric points,were significantly different.The secondary mechanisms of ZxOSCAs and ZxbZIPs were dominated by alpha helix and random coil,while ZxNACs and ZxWRKYs were dominated by random coil.The subcellular localization of ZxNACs,ZxbZIPs and ZxWRKYs was mostly located in the nucleus.3.Every gene family members of Zygophyllum xanthoxylon have the unique conservative structural domain of this family,and the composition of the conservative motif is related to the classification of each family members,which can provide a stronger basis for the classification.4.Except that ZxbZIP18 has no tyrosine phosphorylation site,and ZxWRKY35 has no threonine phosphorylation site,other members of the four families have serine,threonine and tyrosine phosphorylation sites.5.The OSCA gene family members all have transmembrane domains,and their functions may be the same as those of OSCA in Arabidopsis thaliana.6.Construction VIGS vector pTRV2-ZxPDS and pTRV2-ZxOSCA1.3.To sum up,these studies predicted the structure and function of four gene families in Zygophyllum xanthoxylon,providing a basis for further study on the function of drought-resistant genes in Zygophyllum xanthoxylon.
