ObjectiveBased on bioinformatics,this study analyzes the expression and clinical prognosis of sperm associated antigen 5(sperm associated antigen 5,SPAG5)in bladder cancer,and validates it in clinical pathological specimens,aiming to personalize and refine bladder cancer in the future Diagnosis and treatment provide new targets.MethodsAnalyze the m RNA expression of SPAG5 in different bladder cancer data sets by searching the Oncomine database.Download the clinicopathological information of SPAG5 in GEO database GSE13507,and analyze the correlation between SPAG5 gene expression and clinicopathological characteristics by chi-square test.Kaplan-meier and Log-rank perform survival analysis.Identify the type and frequency of SPAG5 gene mutations in bladder cancer through the c Bio Portal website,and analyze the relationship between gene mutations and prognosis.The correlation between SPAG5 and bladder tumor immune cell infiltration was analyzed through the TIMER website.Use Linked Omics and DAVID website to analyze SPAG5 related expression genes and KEGG pathway,verify the protein expression level of SPAG5 in bladder cancer and adjacent tissues by immunohistochemistry,and analyze the relationship with clinicopathological characteristics.ResultsThe m RNA expression level of SPAG5 in bladder cancer tissues is significantly higher than that in normal tissues,and it has nothing to do with age and M staging,but it is related to gender,aggressiveness,tumor grade,T staging,and N staging.The specific survival rate and overall survival rate of patients with high SPAG5 expression are lower than those with low expression.Up-regulation of m RNA is the most common variant of SPAG5 in bladder cancer.SPAG5 is positively correlated with cell purity,expression of CD8+ T cells,and dendritic cells,and it may be involved in tumor immune infiltration.SPAG5 related genes are mainly related to cell cycle,oocyte division,chemokine signal pathway,Gn RH signal pathway,homologous recombination,P53 signal and so on.Immunohistochemical verification showed that the expression level of SPAG5 in bladder cancer tissue was significantly higher than that in adjacent tissues,and it was related to aggressiveness,T staging,but not related to gender,age,disease grade,tumor size,and N staging(P>0.05).ConclusionsOverexpression of SPAG5 is an important factor for the poor prognosis of bladder cancer.SPAG5 has gene mutations in bladder cancer and may be involved in tumor immune infiltration.
