In December 2019,COVID-19 broke out in Wuhan,China,and then quickly spread to the world,attracting great attention at home and abroad.As of the completion of this paper,there is still a lack of effective methods for the preventionand treatment of the virus.Therefore,research on coronaviruses,especially basic research,still needs to be strengthened.This study provides a theoretical basis for the prevention and treatment of COVID-19 and other coronaviruses that may break out in the future through the comprehensive analysis of viral genome and other data,and also provides new directions and ideas for the research on coronaviruses and even the entire nidovirus.The applicant intends to solve two key scientific problems in this research:(1)the main genomic characteristics of SARS-Co V-2 and its role in transmission and outbreak;(2)whether the regulatory region of the jumping transcription and the genome recombination regions of SARS-Co V-2 are similar in nucleic acid sequence.The conclusions and main contributions of this study:(1)Recombination is the most fundamental factor that leads to the strong cross-species transmission ability,wide host adaptability and repeated outbreaks of the coronavirus;(2)The jumping transcription and recombination of coronaviruses have the same molecular mechanism which inevitably cause the outbreak of coronavirus;(3)The optimization of a single factor is not enough to cause a large outbreak of the virus.Only through recombination to combine optimized single factors in different virus strains can a super virus strain with strong transmission ability and wide host adaptability be produced;(4)The WIV1 virus strain without the enhanced ORF8 and the Ra TG13 virus strain without the Furin protease cleavage site at the junction of S1/S2 may have contributed to SARS and COVID-19,respectively,but not their immediate ancestors,and pangolins are not an intermediate host of SARS-Co V-2;(5)It has been confirmed for the first time in the world that the TRS motifs of all coronaviruses(or even the entire nidovirus)are nucleic acid sequences starting with A and having a length of 6-8bp and the recombination region of coronaviruses is also a nucleic acid sequence starting with A,which completely solves the problem of TRS motif and recombination identification;(6)The TRS motifs of all coronaviruses have been re-identified.
