With the further development of brain and nervous system research,the neural mechanism of cerebellar learning and memory has been well known.Recent studies have shown that Purkinje cells are the only output neurons in the cerebellar cortex,which play a key role in sensory perception and motor control.However,the role of Purkinje cells in cerebellar information processing remain poorly understood.Therefore,the complex spike response mechanism of cerebellar Purkinje cells is studied in this thesis.The main contents of the thesis include:(1)Firstly,based on the existing physiological experimental data of Purkinje cells and a variety of complex spike response waveforms,a complex spike recognition and extraction method is proposed in this thesis.In addition,by setting different parameters of synaptic stimulation of climbing fibers on Purkinje cells,the mechanism of complex spike response is analyzed.The results show that the number,distribution area and maximum conductance of synapses in climbing fiber have important effects on the duration of complex spike response and spikelet peak and number.(2)In order to better explore the mechanism of complex spike response in Purkinje cells,the correlation between complex spike response and different ionic currents is further investigated.Firstly,the correlation between several main ion currents and complex spikes is analyzed via the distribution characteristics of ion channels in Purkinje cells.Secondly,this thesis studies how the cellular voltage modulates the duration and pause of complex spikes.Finally,based on the output of simple spikes from cells in combination with the instantaneous burst characteristics of complex spikes,the modulation effect of complex spikes on simple spikes is analyzed.The results show that the complex spikes are promoted or inhibited by different ion currents,and the pause duration after the complex spikes is positively correlated with the cell polarization state.The emission frequency of simple spikes will be inhibited in a short time when the increasement of the number and duration of spikelet of complex spikes.In summary,this thesis focuses on the complex spike response mechanism of cerebellar Purkinje cells,proposes a complex spike recognition and extraction method,and then better reveals the electrical response principle of Purkinje cells through experimental analysis of climbing fiber synapses and ion channels.
