This thesis is created under the social background of the increasing demand for nucleic acid detection.It aims to realize the automation and simplification of nucleic acid detection process,and is expected to popularize the whole process of nucleic acid detection to grass-roots medical units on a large scale.Detection of nucleic acids is an important basis for the detection of certain diseases,but the target nucleic acids usually exist in body fluids at a very low concentration,and the components of these body fluids are often very complex,which interferes with the detection of target nucleic acids.It can accurately extract and detect nucleic acids from complex samples,and the operation is automatic and simple.At the same time,the highly integrated and automatic operation of the detection platform can provide a very good guarantee for the safety of detection personnel.Based on the new idea provided by microfluidic chip for nucleic acid detection of complex samples,the software program is based on STM32F407 main control chip,uses UCOSIII as the embedded control program of multi-step motor of operating system,and uses Devon serial port screen control screen to make human-computer interaction.There are three main steps in the detection experiment of this subject.Firstly,preprocess the sample,The cells containing the target nucleic acid in a complex sample are separated and purified with other cells by microfluidic chip,and then PCR amplification is completed by self-developed consumables.Finally,a self-designed detection device is designed to verify and analyze the nucleic acid to prove the correctness of the device.Finally,the function and effect of the equipment are verified,the separation and detection experiments of complex samples are completed,the feasibility of the device is proved,and the full-automatic equipment integrating separation,purification and PCR amplification is realized.In the test,the complex sample processing device can realize the envisaged functional characteristics of high sample separation efficiency,low equipment cost,easy construction,low detection difficulty and convenient operation.The social demand for nucleic acid detection is gradually increasing.The complex system detection system can be used to solve social needs and practical problems.
