In recent years, photoacoustic imaging technology with the high resolution of optical imaging technology and the high penetration depth of acoustic imaging technology, has been widely used in the biomedical field. In this study, the imaging of normal liver tissue and diseased liver tissue in mice was studied based on photoacoustic imaging technology, providing new ideas for the imaging diagnosis of liver diseases. In addition, the photoacoustic microscopy system based on transparent transducer is proposed, which can improve the optical path module of traditional photoacoustic imaging system,so that the system can facilitate the development trend of miniaturization and take into account the wide imaging field and fast imaging speed, and has a new development opportunity in the biomedical field. The photoacoustic microscopy system based on transparent ultrasonic transducer shows that the homemade 7 MHz ultrasonic transducer has good performance on transmittance, electromechanical coupling coefficient and pulse-echo response. The performance of high resolution, wide imaging field and fast imaging speed was verified in experiments of carbon fiber and mouse ear, showing that the system has a high application prospect in the biomedical field. The results show that photoacoustic imaging technology can clearly show the main blood vessels around the liver and the overall morphology of the liver. In normal liver tissue, photoacoustic imaging technology can observe the complete liver lobule structure, but also detect the liver surface oxygen metabolism and other parameters. The liver tissue was observed under the disease model, and it was found that the hepatic lobule structure was damaged, presenting an incomplete structure, and oxygen saturation and other parameters decreased significantly,in some model mice with severe disease, the liver surface showed a ring structure or the liver surface was blurred to no obvious structure. This result has a good correlation with the staining result of biological tissue.High-resolution photoacoustic imaging technology can realize the imaging of small animals in vivo and observe the microscopic changes of liver and other tissues. It can be used as a complementary method for conventional imaging diagnosis of liver diseases, and is conducive to providing a new research method for imaging diagnosis of liver and other tissue diseases,so as to carry out further research on related diseases.
