Poly malic acid(PMLA)is a biodegradation,chemical derivative,water solubility,non-toxicity,non-immunogenicity biopolymeric product.So PMLA has extensive application in the areas of food,cosmetics,pharmacy and medical industry.Currently,organic solvents are used for the precipitation of PMLA,resulting in great harm to environment and high energy consumption.In this paper,full hydration extraction process was studied,which was low harm to environment,low production cost and ambient condition.Here are main conclusions of this article:In this article,plate and frame filter was selected to remove fungus.Determined by single factor and orthogonal experiments,the best conditions for the filter cloth aperture of 1200 mesh,diatomite addition amount of 1.4%(w/v),the concentration of PMLA solution is 25 g/L.In this condition,the fungus removal rate of 99.69%,the PMLA yield of 94.17%.Decoloring process was respectively dealt with carbon and H2O2,depending on single factor and orthogonal experiments.The decolorization effect,yield and molecular weight of PMLA were used as indexes,carbon decolorization was chosen to continue,the best conditions for the type of E(ZC-A),activated carbon addition of 1.2%(w/v),PMLA solution pH of 4.0,heating time of 30 min,heating temperature of 45?.In this condition,the decoloring rate of 91.4%,the PMLA yield of 87.73%.Membrane separation technology was used to remove macromolecular material.Firstly,select suitable ultrafiltration membrane aperture size(50 kDa),and optimized by single factor and orthogonal experiment,the best conditions for the concentration of PMLA solution is 25 g/L,solution temperature is 50?,the operating pressure is 0.14 MPa,the time of add water elution is 4 times.In this condition,PMLA transmittance 84.76%,the polysaccharide yield of 66.83%.Pullulanase was used to further remove the polysaccharide.Depending on single factor and orthogonal experiments,the best conditions for PMLA solution pH of 6.0,heating temperature of 55?,the addition of 1.5%(v/v)(polysaccharide of PMLA solution was 10.17 g/L),heating time of 80 min,in this condition,the PMLA yield of 79.83%,reducing sugar content of 7.96 g/L.The method of thermal denaturation protein removal was selected.Determined by single factor and orthogonal experiments,the best conditions for pH of 7.5,heating temperature of 90?,time of 30 min.In this condition,the protein removal rate of 89.93%,the PMLA yield of 77.80%.Membrane separation technology was also used to remove small molecular material from PMLA solution.Firstly,select suitable ultrafiltration membrane aperture size(1 kDa),and optimized by single factor and orthogonal experiment,the best conditions for the concentration of PMLA solution is 30 g/L,PMLA solution temperature is 50? the operating pressure is 0.12 MPa,the time of elution is 4 times.In this condition,the ion removal rate of 76.8%,the polysaccharide loss rate of 85.16%,the PMLA yield of 70.91%.To identificate and analyse the PMLA,this annlyse conclude that the PMLA from our study is poly malic acid.The preparation of novel biodegradable and edible nanocapsule based on self-assemble of PMLA and Chitosan(CS)under normal temperature and pressure was described in this part.Depend on response surface optimization experiments,the best conditions for pH value of PMLA is 6.6,mass concentration of CS is 0.9 g/L,volume of PMLA is 4.3 mL.The Z-Ave of the nanocapsules prepared under the best contidions is 126.1 nm.
