A novel silica-particle-supported polymer monolithic solid phase with multi-mechanism of interactions was prepared for capillary liquid chromatography by in situ coating 5 ?m bare silica particles with the copolymers of glycidyl methacrylate and pentaerythritol triacrylate,and further modifying with L-lysine inside a 150 ?m i.d.fused capillary(the prepared column was shorted as “L-column” hereafter).The conditions for preparation of the L-column were investigated based on SEM images and pressure-vs-flowrate curves.The established conditions were as follows: porogenic solvent: methanol/cyclohexanol=1:15(v/v);total monomer concentration,T=25%,C%=30%;polymerization reaction time,5 h;polymerization temperature,60?.Then,the resulting column was characterized in terms of matrix morphology,swelling-shrinking behavior,permeability,mechanical stability,hydrophilicity,binding capacity,preparation reproducibility and column efficiency.The results demonstrated that the L-column was superior to its organic counterpart in pore morphology,mechanic stability,binding capacity and the resistance to swelling-shrinking,and was comparable to the particle packed columns in permeability.In addition,the L-column was prepared easily and conveniently under mild reaction conditions and can be cut in any desired length.Finally,chromatographic performance of the L-column was examined by separations of metal cations,inorganic anions,amines,nucleotides and amino acids.The influence of the composition of mobile phase,p H value,ionic strength and content of ACN on chromatographic separations was investigated.The results demonstrated that the L-column can be applied for chromatographic separations based on ion exchange,hydrophilic,ligand exchange interactions or the mixed mode of these interactions.
