Objective:In this paper,different methods of deodorization and decolorization were used to extract the skin of Rana chensinensis.The main ingredients of the Rana chensinensis skin and the traditional Chinese medicine CCMM are to develop the paste with whitening and moisturizing effect.Select the best preparation,optimizting the process and establishing primary quality standard with discussing the efficacy of the paste.Methods:1.The skin of Rana chensinensis has been cut,deodorized,decolored,vacuum dried(80?,3 h),smashed(200 mesh)and with getting the crude extract.The effects of tea deodorization,?-CD deodorization,ozone deodorizationan,compound deodorization on deodorization were studied by 1-pentene-3-alcohol content,thiobarbituric acid detection and sensory evaluation;The effect of hydrogen peroxide,activated carbon and macroporous adsorption resin on the decolorization effect were studied by the calculation of decolorization rate and loss rate;The Fourier infrared spectrometer(FTIR)is used to analyze the active ingredients in the skin of Rana chensinensis.2.The appearance score and moisturizing evaluation of the paste with main material were evaluated by investigating the crude extract of Rana chensinensis skin(1.0?1.5?2.0%)and CCMM(2.0?3.0?4.0%).Optimizting the matrix formulation of the paste by the single factor experiments and response surface analysis methods.3.The content of hyaluronic acid in paste was determined by Bitter-Muir carbazole.The content of collagen was determined by Worssner colorimetry and the content of total saponins was determined by colorimetric method.The stability of the paste was determined by the appearance,cold resistance,heat resistance,centrifuge test,emulsification stability and pH.4.The whitening property of paste was explored by measuring the inhibition rate of tyrosinase.Determination of moisture absorption and moisture content of paste in vitro;Moisturizing effect of mouse skin in short-term tests(180 min)and long-term trials(15 d).Results:1.The content of 1-pentene-3-alcohol in Rana chensinensis skin decreased from 10.82%to 0.42%by gas chromatography after compound deodorization.The content of thiobarbituric acid and the fishy value were the lowest.The trend of deodorization effect was compound deodorization>tea deodorization>?-cyclodextrin decontamination>ozone decontamination.The trend of deodorization effect was H2O2>macroporous adsorption resin>activated carbon.The active ingredients including hyaluronic acid and collagen were analyzed by FTIR.2.The compound Rana chensinensis skin whitening moisturizing paste(CRCP)with the best appearance and the best moisturizing effect was formulated by using 1.5%crude extract of Rana chensinensis skin and 3.0%CCMM crude extract.The highest comprehensive score(9.34)by using 8%stearic acid,1.5%stearic acid glyceride,12%glycerol.3.The content of hyaluronic acid in CRCP was 0.19 mg/g,the content of collagen was 10.26 mg/g,and the total saponin content was 1.14 mg/g.4.The inhibition rate of tyrosinase activity of CRCP was(39.28±0.53%)with good whitening effect,and it was close to(42.77±0.48%)of market whitening moisturizing paste.5.The CRCP and the positive control group paste have significant difference compared with the control group(p<0.05),the CRCP has no significant difference compared with the positive control group and hyaluronic acid paste group(p>0.05)in the short term(180 min)moisture retention experiment.The CRCP has significant diffrence compared with the positive control group and hyaluronic acid paste group(p<0.05),and have extremely significant difference compared with the control group and CCMM paste group(p<0.01)in the long term(10,15 d)moisture retention experiment.ConclusionThe best effect Rana chensinensis skin is decomposed by composite method and decolorized by H2O2.CRCP with the best appearance and the best moisturizing effect was formulated by using 1.5%crude extract of Rana chensinensis skin and 3%CCMM crude extract.And with the the highest score by using 8%stearic acid,1.5%stearic acid glyceride,12%glycerol.The remarkable whitening and moisturizing effects of CRCP were verified by tyrosinase inhibition,in vitro hygroscopicity and moisturization tests,and smearing of mouse skin.
