Characteristics of gut microbiota and host metabolism are hypothesized to be associated with constipated status,but the regulation mechanism is not fully understood.Thus,the current study investigates effect of constipation symptoms on gut functionality following the modulation of gut microbiota and metabolites via KOS intervention.Effect of alteration of gut microbiota and metabolites on constipation symptoms induced by KOS was investigated using a constipated rats model,which was given diphenoxylate by gavage.The content studyed in this paper are as followes:serum biochemical parameters,fecal short-chain fatty acids(SCFAs),histological analysis of colon tissues,gene expression related to tight junction protein and water transport,composition and diversity of gut microbiota.The data of serum biochemical parameters revealed that blood metabolism of host was improved significantly after the KOS intervention through inhibited oxidative stress,improved lipid metabolism and insulin resistance,which was reflected by the improvement of T-AOC,activities of SOD and GSH-Px(P<0.05),TC,TG,LDL-C and HOMA-IR(P<0.05).Feces of KOS groups were found to have a significant higher contend of SCFAs in the manner of M-KOS>H-KOS>L-KOS,and the high level of butyrate was associated with the abundant butyrate-producing bacterias(Bacteroidales S24-7 group and Ruminococcaceae).Fuethermore,increased thickness of the mucosal layer,achieved by KOS supplement,was found to improve the gut environment and colonic barrier function possibly through improving the expression of tight junction proteins and aquaporins.Because the data showed that the expresion of Claudin-3,Claudin-4 and ZO-1 were enchanced significantly(P<0.05),while the expression of AQP1,AQP3,AQP4,AQP8,SMIT and TauT were decreased significantly(P<0.05).More importantly,the abundance of gut microbiota was enhanced,which verified by the Alpha diversity indexes and curves.The analysis of Beta diversity enhibited that the structure of gut microbiota was significantly improved,in particular the H-KOS group which was similar to the NC group.And the abundance of Bacteroidales S24-7 group and Ruminococcaceae on the family level were higher after KOS supplement,which was associated with the increased butyrate producing.In conclusion,metabolic disorder and gut barrier integrity were improved in constipation status after KOS intervention.Furthermore,enchanced the diversity and structure of gut microbiota,characterized with enriched Bacteroidales S24-7 group and Ruminococcaceae,attenuated constipation symptom after KOS intervention.
