Xinjiang blend tea(XBT)is a type of health tea,which has a history of more than 1600 years.It was listed in the fourth batch of intangible cultural heritage of autonomous region in 2014.In this experiment,Xinjiang tea was prepared from cardamom,safflower,rose,clove,peppermint,cumin,licorice,ginger,turmeric and Fuzhuan tea.The physical and chemical indexes were determined by national standards.The components of tea aroma were analyzed by GC-MS.Tea polyphenols was used as the processing technology index.The Box-Behnken method was used to optimize the processing technology.The effects of reducing power ability,free radical scavenging activities on ABTS+and DPPH·,as well as cytoprotective activity on H2O2induced Hep G2 cell injury.Moreover,the immune regulatory capacity of XBT on RAW 264.7 macrophages immune were also examined.Results are as follows:(1)Result indicated that the XBT contained 5.92%of polyphenol,7.92%of soluble sugar,6.29%of crude fat,16.381%of crude protein,12.67%of amino acids,20.97%of crude fiber,8.45%of ash,and11.14%of moisture.Moreover,the yield for water extract was up to 23.47%.Apart from that,a total of 17amino acids were detected,which contains 36.84%essential amino acids.The amino acids analysis showed that the content of Met+Cys was 30.35%of recommendation value,which were the first limiting amino acids.Last but not the least,GC-MS analysis illustrated that the phenols were the highest(41.91%),followed by alcohols(11.48%),hydrocarbons(8.87%),ketones(8.4%),esters(3.37%),acids(2.01%),and aldehydes(0.7%)for about 72 fragrant components.(2)The optimal extraction conditions were determined as:extraction time 24 min,extraction temperature 85.5?,liquid to material ratio 54 g/g.Under optimization condition,the yield of polyphenols was 46.86±0.814 mg/g.The IC50of ABTS+and DPPH·were as follows:30.837 mg/m L and 13.399mg/m L,Hep G2 cells have a significant protective effect against oxidative damage,suggesting that XMT possessed strong reducing power and total antioxidant capability.(3)The experimental results showed XBT did not affect the cell vitality at low concentration.Furthermore,XBT can significantly stimulate RAW 264.7 cell to release NO.The result of q RT-PCR analysis indicated that XBT also enhanced inducible nitric oxide synthase(i NOS),cyclooxygenase-2(COX-2),TNF-?and IL-1?expression.Further analyses demonstrated that WBP rapidly activated phosphorylated mitogen-activated protein kinase(MAPK)and the transcriptional activities of activator protein-1(AP-1)and nuclear factor(NF)-?B.Taken together,these results suggested that XBT can improve immunity by enhancing immune function.
