Heart valve diseases are a common and frequently-occurring disease,and cardiac valve replacement is the most effective surgical treatment for end-stage heart valve diseases.At present,the main substitute valves used in clinic are mechanical heart valves and bioprosthetic heart valves.However,they are not ideal substitutes for heart valves.As a new type of material,polymer heart valves solve some existing problems,but there are also some unsolved problems such as hydrodynamics.In order to solve the existing problems,SF-PEG artificial heart valve composite material was prepared by electrospinning technology using silk fibroin(SF)as raw material.The results show that anisotropic SF-PEG composites can be obtained by optimizing the process parameters and combining UV crosslinking.The elastic modulus of SF-PEG composites in the parallel direction is 9.96 ± 1.12 MPa and the elastic modulus in the perpendicular direction is 3.12±0.30 MPa.The mechanical properties of SF-PEG composites meet the demand of human heart valves.Furthermore,dynamic mechanical thermal analysis and stress relaxation tests show that SF-PEG composites have good energy absorption and diffusion ability,which have the advantage of dealing with complex blood environment.In the enzymatic degradation experiment and the dynamic calcification experiment,SF-PEG composites have the ability of anti-enzymolysis and anti-calcification due to the fact that the special grid structure of PEG hydrogel has the ability of anti-fouling.At the same time,SF-PEG cardiac valve stent was fabricated by 3D printing resin heart valve stent.Through the curve analysis of the Vivitro Pulsation Platform,SF-PEG heart valve scaffold has an effective opening area of 2.6 cm2 and a regurge of 20%,which conforms to the ISO 5840 standard.It proves that SF-PEG heart valve scaffold has good hydrodynamic characteristics and has the potential to become a new prosthetic heart valve scaffold.
