With the rapid development of electronic technique and bioscience research,nanopore single molecule analytical techniques have attracted increasing attention all over the world in the last decades.The resistive pulses caused by molecule translocation through a single nanopore have been considered as a powerful detection means of single molecule events.Due to nanopore single molecule analysis involved small current(pA level)and fast transition(ms or ?s scale),this method need construction of the single nano interface and high sensitive electronic detection.Nevertheless,the control of environmental electro-magnetic disturbance as well as noises from various sources is also critical.In the first chapter,the sorts of single molecule analysis and characteristic of biological/solid-state nanopore were introduced.In addition,present developing status and typical problem were exposited.The fundamental composition and function of detection system were summarized in detail.In the second chapter,baseline current was measured in different gains by EPC 10 system for acquiring the optimal gain value.In addition,a compatible analog-digital converter was selected and the whole detection system was established based on current amplifier maded by Long's research group in ECUST.Through the comparison for electrical noise(peak-to-peak value,RMS and power spectrum),this system met the requirement of single molecule detection.In the third chapter,a simply micro-nano interface was directly constructed by PTFE film and glass capillary and successfully applied to support lipid bilayer.Preliminary experiment result showed that it is feasible for nanopore single molecule analysis to integrate the micro-nano interface with detection system.In this work,nanopore single molecule analysis detection system which could be used in common laboratory was established based on domestic current amplifier.Through the measurement of current signal pulse caused by ssDNA transition,performance of this system is up to standard of single molecule analytical technique,such as sample frequency,sensitivity and noise level.This system will facilitate domestic single molecule detection system of technological progress in some degree.
