In recent years,there are,many researches on the application of enzyme in leather processing at home and abroad.Protease,lipase and glucosase can degrade fat,globular protein and proteoglycan to remove fibrous interstitium.If the combination of protease,lipase and glucosase is adopted,the better effect may be achieved.Therefore,alkaline protease JRH,glucosase TH and alkaline lipase JZ were selected in this paper.The formula of the enzymatic compound was determined by taking the total protein concentration,the ratio of collagen to the total protein and the degreasing rate as the indexes.The enzymatic compound was used in the degreasing process of pig skin.The histological structure of degreased pigskin was compared with that of undegreased pigskin.The change of physical and mechanical properties of pigskin was studied.The synergistic mechanism of enzymatic compound in pig skin degreasing was discussed.The enzymatic compound was used in the degreasing process of pig skin,cattle skin and sheep skin.An environmental impact assessment of waste water was carried out.The suitable conditions for the use of enzymes were explored.The results showed that the compound enzyme formula was JRH 20000 U,glucosase TH 40,000 U and alkaline lipase JZ 6000 U per kg of raw skin.The degreasing rate of enzymatic compound on raw pigskin was 82.3%.The total protein concentration in the wastewater was 1570.58 g/mL.The ratio of collagen to total protein was 0.47%.The results showed that the enzymatic compound could remove the fat and collagen fiber stroma effectively.Through the observation of fluorescence microscope and the analysis of the degreasing rate and total protein concentration of the three enzymes,it was found that the alkaline protease JRH,glucosase TH and alkaline lipase JZ had a synergistic degreasing effect.Through optical microscope and scanning electron microscope,it was found that the enzymatic compound had better effect on the removal of fat and the dispersion of collagen fiber than the non-ionic degreasing agent.Compared with the pigskin treated by non-ionic degreasing agent,the pigskin treated by enzymatic compound has better mechanical properties.The enzymatic compound also has good degreasing effect on sheep skin and cattle skin.Compared with the wastewater produced by non-ionic degreasing agent degreasing process,the wastewater produced by enzymatic compound degreasing process has less environmental pollution and is more biodegradable.
