Determination Of Ribavirin And Amantadine In Poultry Meat By Mass Spectrometry

This article is done by 60 mg/kg.b.w.d fed samples of white-feathered chicken ribavirin and amantamine.Under optimization conditions for detection,a linear relationship occurs when the mass concentration of amantlamine and ribavirin is within the interval of 0.50?200?g/L.After sampling and testing,the elimination curve of ribavirine and amantlamine is obtained,and the elimination rules of limavirine in chicken tissues are as follows:(1)The residues of ribavirine and amantanine in different chicken tissues in the early withdrawal were directly proportional to the administration dose.ribavirin and amantanine remained higher in 4 h,tissues after cessation,both higher in the kidney tissue than other tissues.(2)After stopping medication,amantanine in the liver and kidneys remained longer compared to chicken breast tissue;the dose was not correlated with the residue of ribavirin in the tissue,and was metabolized completely after withdrawal.The determination of ribavirin and amantanine residues in chicken,its liver and kidney tissues by high resolution mass spectrometry,and its metabolic rules.In the case of positive ions,ribavirine and amantanine have high response values,the experiment with positive ion full scan.Chicken tissue samples were extracted using trifluoroacetic acid of 1% volume ratio-methanol solution,purified by Qu ECh ERS method and PCX solid phase extraction column,and showed that amantamine had a good linear relationship with ribavirin in the range of 0.50?200 ?g/L.The limits of detection for the two target analytes were 0.30 ?g/kg and 0.15 ?g/kg,respectively,and the lower limits of quantification for the two drugs were 1.00?g/kg and 0.50 ?g/kg,respectively,and the RSDs of the samples were in the range of 6.0%-10.7%.Under optimized chromatography and mass spectrometry,experiments can be conducted in eluting liquid,flow phase and solid phase extraction column.Finally,with negative water and blank chicken,liver and kidney matrix,the added target compound is verified by Q-TOF mass spectrometry screening: the measured material can obtain certain energy through Q-TOF mass spectrometry to divide it into fragmented charged particles based on certain rules.In this way,the detector can calculate the mass core ratio of relevant ions,and draw the corresponding mass spectromegram based on the corresponding order.By comparing the test results,the precise molecular weight of the suspected target drug metabolites was extracted to provide data support for the comprehensive evaluation of drug residues through the landmark metabolites.
