Indole and its derivatives are important classes of heterocyclic compounds,which are widely present in a large number of natural products and bioactive molecules,and are widely used in materials science and pharmaceutical chemistry.In recent decades,the efficient preparation of its derivatives has attracted great attention from chemists,and many synthetic strategies have been developed.In addition,cyano groups are widely found in a variety of natural products and pharmaceutical molecules.Due to its unique chemical properties,cyanide can be easily converted into a variety of organic groups,such as aldehydes,ketones,carboxylic acids,amines,and amides.Therefore,many chemists devote themselves to introducing cyanide into indole compounds,which once became a hot spot in heterocyclic synthesis for a time,and related research results have been greatly developed and applied.As we continue to focus on tandem catalysis and the synthesis of heterocyclic compounds,we herein report a novel one-pot method for the synthesis of 3-cyanoindole,This method can synthesize 3-cyanoindole compounds with N-protected or free-NH unit.The yield is good to excellent.At the same time,it has the characteristics of simple operation and mild conditions.The main content of this paper includes the following two parts:(1)A brief overview of the research progress in the synthesis of transition metal-catalyzed synthesis of indoles in recent years.(2)A new palladium-catalyzed method for the synthesis of 3-cyanoindole from obromoaniline and 4-(4,4,5,5-Tetramethyl-1,3,2-dioxaborolan-2-yl)isoxazole by tandem reaction was studied.Based on this research strategy,not only the simple and effective preparation of 3-cyanoindole has been completed,a variety of heterocyclic molecules have been constructed,but the effective synthesis of a variety of pharmaceutical and pesticide molecules have also been achieved.
