Resveratrol(resveratrol,Res)is a natural non-flavonoid polyphenol compound with anti-cancer,antibacterial,liver protection and other physiological functions.It has become the focus of common concern in the medical and food industries in recent years.The annual output of peanut roots in my country is about 4 million tons,most of which are discarded in the field,a small part is used as feed and fuel with the stems and leaves,and some are underutilized and have low added value.At present,the resveratrol from peanut roots There is less research.This article mainly extracts Res from peanut roots,and uses the macroporous resin adsorption method to separate and purify it,and finally explores the effect of Res on the quality of peanut oil.The main findings are as follows:(1)This article uses water bath,microwave,and enzymatic methods to extract resveratrol(Res)and Res glycosides from peanut roots,and analyzes 19 different varieties using ultra-high performance liquid chromatography(UPLC)under the best extraction process The content of cis and trans Res and cis and trans Res glycosides in peanut roots.The results showed that the crushed mesh number of peanut root had a significant effect on the total content of Res and Res glycosides.The optimized microwave method had the highest total content(4235.59±24.33 mg/kg),followed by ?-glucosidase enzymatic hydrolysis(3914.82±16.07 mg)./kg),water bath method is the lowest(2024.11±12.18 mg/kg),and the content of trans-Res glycoside extracted by microwave method is high,while the content of cis-Res extracted by water bath method and ?-glucosidase enzymatic hydrolysis method is high.Due to the large consumption of organic solvents in the microwave method,the ?-glucosidase enzymatic hydrolysis method was selected for subsequent experiments.(2)Based on the single factor of enzymatic method,design experiments by response surface analysis method,using the extraction amount of resveratrol as an indicator to explore the effect of enzyme addition,temperature,enzymolysis time,and p H on the extraction process,and use the response The surface analysis method optimizes its extraction process,and finally analyzes the difference of resveratrol content in peanut rhizomes from different producing areas.The results showed that when the amount of enzyme added was0.32 mg,the temperature was 70.89?,p H=5.37,and the enzymatic hydrolysis time was 121.71 min,the peanut root powder resveratrol extraction amount could reach 3719.7 mg/kg under these conditions.The total content of Res and Res glycosides in the roots of peanut varieties is different.The total content in the roots of Huayu series peanuts is higher than that of other varieties,and the total content in the roots of Huayu967 peanuts is the highest at 4137.18±19.56 mg/kg,of which cis and trans Res account for 33 % And26%,and cis and trans Res glycosides accounted for 23% and 18%.This extraction process provides a technical basis for the efficient extraction and utilization of Res and Res glycosides from peanut roots.(3)The ?-glucosidase method is used to extract resveratrol from peanut roots,and the obtained extract is subjected to macroporous resin separation and purification.Through dynamic and static adsorption experiments,it is determined that the best separation and purification process of macroporous resin is:Optional The macroporous resin is polyamide,the desorbent is 90% ethanol,the elution rate is 1.5 BV/h,and the purity is 49±0.3% as determined by ultra-high performance liquid chromatography(UPLC).(4)Use magnetic stirring to dissolve resveratrol in peanut oil to study the effect of resveratrol on the quality of peanut oil.The specific process is: 100 mg resveratrol is magnetically stirred at 1000 rpm and50? for 1 h A blank control was carried out with undissolved resveratrol peanut oil,and it was found that the content of trans-resveratrol(576.69 mg/kg)of peanut oil dissolved in trans-resveratrol was greater than the original trans-resveratrol in peanut oil Content(98.96 mg/kg).
