The antioxidant activity of proanthocyanidins has become a research hotspot.Cerasus humilis is a new domestic fruit in recent years,which is rich in proanthocyanidins.In this study,the Cerasus humilis from Zhongwei area of Ningxia was used as the raw material,and the proanthocyanidin from the Cerasus humilis was extracted,purified,analyzed,and the antioxidant properties in vivo and in vitro were studied.The results of the study are as follows:(1)Optimum extraction conditions:In the extraction experiment of Cerasus humilis proanthocyanidins,There were four single studied factors which affected the production of anthocyanin extraction:the effects of extraction time,extraction temperature,ethanol concentration and material-to-liquid ratio and the best extraction conditions of Cerasus humilis proanthocyanidins were obtained.The extraction time was 41 min,the extraction temperature was 45?,the ethanol concentration was 67%,and the material-to-liquid ratio was 1:15g/mL.Under these conditions,the highest extraction rate of proanthocyanidins was obtained,with a content of 24.28%.(2)Purification and composition analysis of proanthocyanidins:Macroporous resin is used to purify the crude proanthocyanidins.Through static adsorption and desorption experiments,AB-8 type macroporous resin was selected for the purification experiment of proanthocyanidins,and it was determined that the adsorption rate of Cerasus humilis proanthocyanidins was the highest under the conditions of pH=4.0 and 25?.In the dynamic adsorption experiment,control the sample flow rate of 75 mL/h,the sample volume of 50 mL,the volume of the eluent 200 mL,the analytical solution is a 60%ethanol solution with a volume of 50 mL and a flow rate of 75 mL/h.The experiment analyzed the components of the purified Cerasus humilis proanthocyanidins.The UPLC-MS/MS method was used to determine and analyze the individual compounds.The test results showed that the monomer components were mainly catechin and epicatechin,followed by tetrameric compounds.Among them,the main content dimer is dimer.(3)In vitro antioxidant capacity study:The experimental results of different free radicals and total antioxidant capacity in vitro show that the order of the free radical scavenging capacity of the three samples is procyanidin standard scavenging ability>procyanidin sample>ascorbic acid,the greater the concentration of proanthocyanidin,the stronger the scavenging ability of free radicals.(4)Study on the protective effect of Oleum proanthocyanidin on mouse liver cells:Carbon tetrachloride was used to establish a liver injury model,and the effect of Oleum proanthocyanidin on liver pathological changes and liver index in mice with liver injury.To detect the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the serum of mice and the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the liver Dialdehyde(MDA)content;the results show that:in the middle dose(50mg/kg)and high dose(100mg/kg)groups,most of the liver tissues are normal,but the liver tissues of the model group have lesions Necrosis,Cerasus humilis proanthocyanidins can reduce the MDA content in mice and inhibit the activity of hydroxyl free radicals;it has a significant effect on improving the vitality of CAT,GSHPX,and SOD;it shows that Cerasus humilis proanthocyanidins can reduce the body's antioxidant capacity by improving the body's antioxidant capacity.Oxidative stress caused by carbon tetrachloride damages liver cells and has a better protective effect on liver damage caused by carbon tetrachloride.
