Cancer is still a major threat to human health,and single chemotherapy can't achieve good effect.Because of its noninvasive and relatively safe therapeutic measure,photothermal therapy has attracted extensive attention in the field of cancer treatment and chemotherapy.However,most of the photothermal agents as chemotherapy drugs by embedding,which results in the reduction of drug loading rate.Here,a novel acylated cyanine dye(MACyanine)with photothermal effect induced by near-infrared light was synthesized.The copolymers poly[2-methyl-2-acrylic acid-2-(2-methoxyethoxy)ethyl-co-polyethylene glycol methyl ether methacrylate-co-phenylboronic acid-co-acrylated cyanine dye](P(MEO2MA-co-OEGMA-co-PBA-co-MACyanine)(P for short)were prepared by reversible addition fracture chain transfer polymerization(RAFT).P-Dex nanodrug delivery system with p H response and near infrared(NIR)light sensitive was constructed by forming reversible borate ester bond between phenylborate and dextran.The anti breast cancer drug Auphen was coated by?-?stacking and electrostatic interaction,dynamic light scattering and scanning electron microscopy showed that the prepared P-Dex-Auphen nano particles were spherical with an average diameter of 130 nm,was suitable for accumulation in tumors through enhanced infiltration and retention effects.The nano system has high drug loading,light stability and high physiological stability.The drug release by nanoparticles showed both p H and NIR stimulated responses.Meanwhile,Auphen blocked the transport of hydrogen peroxide(H2O2)by inhibiting AQP3 water channel protein.Cell scratch and Transwell invasion experiment showed inhibition of cell migration and invasion.In vitro cytotoxicity results showed that nanoparticles could be effectively absorbed by cells.Compared with P-Dex nanoparticles in the blank group,drug loaded nanoparticles showed cytotoxicity with the increase of drug concentration,and the presence of NIR light accelerated the release of drugs and improved the toxicity to cells.The combination of photothermal therapy and chemical therapy,which can produce hyperthermia by photoinduction,realized the effective treatment of breast cancer cells.
