The citrus is rich in China,and it is one of the main fruit in south of China.Its production and planting area are the highest in the world.But during the post-harvest storage and transportation process of citrus fruits,they are vulnerable to be infected of pathogenic fungi and then rotted,which cause severe loss.Therefore,citrus post-harvest fresh preservation technology development has been a hot spot in the industry research.Slow release preservation is one of the most common methods of preserving fresh fruits and vegetables.However,it has been less applied to citrus preservation.There were substantial evidences indicating that the essential oil rich in pinene has broad antimicrobial activity,and the effect of the inhibition to penicillium italicum is obvious.In current study,we intended to take pinene as the main object,studied the preparation technology of sustained-release preparation and its characterization and function on post-harvest citrus fresh preservation,to develop a new solid sustained-release anti-staling agent which is suitable for citrus preservation,and then providing a new avenue of citrus preservation for prolonging storage.The main results were as follows:1 ? The screening of ?-pinene sustained-release preparation preparing process and determination of optimum parametersThe antibacterial effect of ?-CD embedding method,microcapsule method and adsorbent method were compared by using antibacterial test in vitro.It was found the optimum preparation of ?-pinene sustained-release preparation was absorbent method,and the optimum adsorbent was silica gel;The optimum ratio of ?-pinene sustained-release preparation was as follow: the mixing ratio of silica gel and ?-pinene is1:2.662(mass ratio).2?Evaluation of release performance of ?-pinene sustained-release preparations and Its structural characterizationThe MIC of ?-pinene sustained-release preparation on P.Italicum is 4.8 mg/m L in vitro;The sustained-release preparation was stored at room temperature for 7 days,and the release rate was 76%,about accumulated 10 days release completely by gas chromatography.The SEM and FTIR analysis show that the adsorption mechanism of?-pinene sustained-release preparation was due to the special internal pore structure of micro-silica gel,which was physical adsorption.3?Study on the fresh citrus preservation of ?-pinene sustained-release preparationIn vivo experiments,we inoculated the P.Italicum to the citrus,its inhibition rate of is up to 29.49% through the fumigation of the 200 mg/L ?-pinene sustained-release formulations compared with the control of clear water group.Also,the rot rate of fresh citrus dealt with ?-pinene sustained-release preparation was reduced by 26.67% compared to natural storage citrus.In addition,the total phenolics,flavonoids and PAL of the citrus peel at different storage times were measured,and we found that the contents of PAL enzyme activities was always improved,and the contents of total phenolics,flavonoids were improved before 12 days in storage.It shows that,the preparation enhances the biological resistance of citrus peel.Therefore,?-pinene sustained-release preparation can be used for the preservation of the citrus harvest shelf life,with market development prospect as a new solid sustained-release preservative.
