Biomarkers can objectively represent different physiological processes of life,and their detection plays an important role in disease diagnosis and prevention.Although the detection of biomarkers is relatively mature and efficient,its application in clinical diagnosis is still lack of accuracy,sensitivity and specificity in many cases.In this study,we build a variety of nanomateriala functional biosensors to achieve rapid detection and high-throughput detection of disease biomarkers.Our research work is mainly divided into the following three parts.(1)An ITO based electrochemical sensor(ITO/Chi/MWCNTs-Tf/BSA)based on the conductivity and easy modification of MWCNTs was constructed and applied to the detection of transferrin receptor.This method is convenient,fast and low cost.It has high specificity and electrochemical response in the detection.At the same time,its detection limit is 0.106 ng/m L,which is far lower than the traditional ELISA method(75.7 ng/m L),so as to realize the rapid and accurate detection of transferrin receptor.(2)Gold and platinum nanoparticles modified e ITO sensors can be used in the electrochemical detection and fluorescence enhanced detection of biomarkers.When Au:Pt=1:0,its electrochemical response range is1.02×10-6 A?1.98×10-6 A during detecting Tf R of different concentrations from 100 ng/m L to 104ng/m L,and the standard curve is I=-2.41×10-7log[CTf R]+1.99×10-6,the correlation coefficient is R2=0.9982,showing good performance;at the same time,when it is used to enhance fluorescence,the fluorescence intensity per unit area is about 62±2,with obvious enhancement effect than unmodified ITO conductive glass.The design of double platform is helpful to improve the detection accuracy of biomarkers.(3)LCM and MALDI MS were combined to detect the single cell metabonomics of cervical cancer.PCA and OPLS-DA models were used to analyze the mass spectrometry results.The results showed that the difference between the two groups was significant(OPLS-DA:R2=0.771),and its AUC was 0.8008.At the same time,we screened 10 potential biomarkers based on S-plot and VIP value.We provide a very important technical platform for exploring the molecular biological mechanism of cervical cancer and early screening of cervical cancer.
