Bee pollen is made by honeybees collecting male reproductive cells in plant stamens and adding their own glandular secretions.FBP is one of the highest yield bee pollens in China and has been used as a health food since ancient times.Its chemical composition and biological activity have attracted extensive attention from scholars.In this paper,the chemical constituents of FBPE were identified by HPLC,the TPC,TFC and antioxidant activity of FBPE in vitro were measured.Based on these,HFD-STZ induced T2DM mice model used to study the hypoglycemic function,regulation effect on intestinal flora and fatty acid metabolism in liver tissue and serum of FBP.This paper is divided into 6 chapters and the main research results are as follows:1.FBP was extracted by percolation at room temperature and chemical composition were identified by HPLC.The TPC,TFC,the antioxidant activity in vitro and the protective effect on DNA damage were studied.In this study,8 phenolic compounds were identified from FBPE,including luteolin,resveratrol,kaempferol,etc.The results showed that the TPC was18.59±1.39 g GAE/kg,the TFC was 16.35±0.09 g RE/kg,and the results of antioxidant activity in vitro showed the DPPH free radical scavenging ability was 11.01 mg/m L(IC50 value),the Fe2+chelating force was 32.84±1.49 mg EDTA-2Na/g,and performed a protective effect on·OH-induced oxidative damage of p BR322 plasmid DNA.2.The hypoglycemic function of FBPE was studied in HFD-STZ induced T2DM mice.Results showed that a continuous intragastric administration FBPE after 8 weeks,FBG,insulin tolerance,serum and hepatic lipid accumulation were reduced,insulin sensitivity was improved,oxidative damage and inflammation were alleviated,and AKT and PI3K mRNA expression were up-regulation.Moreover,the symptom such as liver cell hypertrophy,hepatic fat accumulation and inflammatory cells infiltration were alleviated,and epididymal fat injury was improved,too.3.The regulation effect of FBPE on intestinal microflora imbalance in HFD-STZ induced T2DM mice was studied by 16sRNA sequencing method.The results showed that FBPE can effectively reduce the relative abundance of SMB53,increase the relative abundance of Coprococcus,Ruminococcus and Turicibacter on genus level,effectively adjust the intestinal microflora dysregulation in HFD-STZ induced T2DM mice.4.The effect of FBPE on fatty acid metabolism in liver tissue and serum of HFD-STZ induced T2DM mice was studied by GC-MS analysis.The results showed that FBPE could significantly increase the relative contents of C16:0 and C18:3n-6 fatty acids,and decrease the relative contents of C17:0,C20:0 and C20:3n-6 fatty acids in liver of T2DM mice.It could significantly increase the relative contents of C15:0,C17:1,C18:3n-3,C20:1,C20:3n-6 and C20:4n6 in serum fatty acids of T2DM mice,significantly reduce the relative contents of C14:0,C16:0,C18:0 and C22:1n-9 fatty acids,significantly increase the relative contents of MUFAs and PUFAs in serum,reduce the relative contents of SFAs,and increase the relative contents of n-3 PUFAs and n-6 PUFAs(P<0.05).FBPE could regulate the changes of fatty acid profile in serum and liver of HFD-STZ induced T2DM mice effectively.
