Soy molasses is a derivative by-product obtained from the industrial production of soybean protein concentrate.It contains substances required for fermentation by a variety of microorganisms,and most of which contains soybean oligosaccharides.It is an ideal fermentation substrate for microbial fermentation.Starting from the composition of soybean molasses,this research explores the fermentation and utilization of soybean molasses by yeast and explores the optimal technology for extracting ribonucleic acid from the obtained bacteria,providing ideas for the comprehensive utilization of soybean molasses.Through the analysis and testing of the main components of soybean molasses,the basic components of molasses mainly include 46.45% of sugars,2.4% crude protein,3.5%fat and 3.73% of ash.Among them,sugars are mainly 28.93% sucrose and 13.54%stachyose,Raffinose 2.86%.According to the types of oligosaccharides contained in molasses,four different yeast strains were selected for fermentation comparison,and finally it was determined that Pichia pastoris was used for fermentation experiment on soybean molasses.After 24 hours of fermented soybean molasses,the sucrose content and stachyose content in the molasses were reduced by 89.34% and 78.48%,respectively,and the raffinose content was reduced by 15.37%.Through the results of single-factor experiments,select the sugar content,ammonium sulfate addition amount,and potassium dihydrogen phosphate addition amount for response surface test,and finally determined the best soybean molasses medium composition: sugar content 9.398 °Brix,ammonium sulfate 0.118%,diphosphate Potassium hydrogen is 0.054%,and the predicted value is 12.63 g/L.After the correction,the sugar content is 9.4 °Brix,ammonium sulfate 0.12%,and potassium dihydrogen phosphate 0.05 %;the dry weight of the bacteria obtained by culturing Pichia pastoris with this medium is 1.35 times that before optimization.The optimal fermentation conditions were determined by single factor experiment: inoculation amount 6%,culture temperature28 °C,shaker rotation speed 180 r/min,natural p H.The process of extracting ribonucleic acid from Pichia pastoris was studied.Use the optimal medium and fermentation conditions to cultivate and collect the bacteria for the ribonucleic acid extraction test.The main factors for extracting ribonucleic acid by salt method: holding time,holding temperature,bacterial solution concentration and Na Cl concentration are single factor test,and then orthogonal test is carried out on the basis of single factor test,and finally the best extraction process is: holding time 5 h,the concentration of bacteria solution is 8%,the salt concentration is 8%,and the holding temperature is 90 ?.The extraction rate and leaching rate under this condition were5.93% and 92.61%,respectively.This research explores the possibility of using soybean molasses as a fermentation substrate,explores new ways to use soybean molasses,reduces environmental pressure on enterprises,and reduces waste emissions;at the same time,it also provides a cheaper and more accessible fermentation substrate for the yeast industry.Turn waste into treasure and reduce production costs.
