The Surface enhanced fluorescence(SEF)refers to the phenomenon that the fluorescence emission intensity of the fluorescent molecules distributed on the surface of noble metal or near its sol increases greatly compared with that of the free state.At present,the research on fluorescent enhanced substrate has become the focus,but there are many shortcomings in many nanostructure fluorescent enhanced substrates,such as complex synthesis steps and high cost,which will hinder the application and development of surface enhanced fluorescence.Based on this,in this paper,we construct metal materials on the surface of natural substrates with large-scale nanostructures to obtain surface enhanced fluorescence substrates and study their optical properties.The specific research contents are as follows:(1)Copper and silver films were modified on micro papillary structure and their fluorescence intensity was studied.Lotus leaf with micro papillary structure was selected as the substrate template.Copper and silver were sputtered onto the substrate by magnetron sputtering,and the effects of the two metals on the fluorescence signal were compared.Three dimensional finite difference time domain method is used to simulate the electromagnetic field enhancement of two kinds of metals,and a preliminary study is made on the enhancement of fluorescence signal on the biological substrate template.(2)The fluorescence enhancement properties of nano columnar structure and the practical properties of the substrate were studied.Dragonfly wings with nano columnar structure were used as template to study the effect of nano silver structure on fluorescence signal.High intensity “hot spot”is adjusted on the surface of metal substrate,and the enhancement of fluorescence signal is analyzed from the mechanism.Meanwhile,the enhancement factor is modified.Finally,the stability,reproducibility and reusability of the optimal substrate are studied.(3)The fluorescence enhancement performance of nanoscale grating structure was studied,and its application in detection technology was explored.The CD/DVD disc with nano-grating structure is used as the substrate template to study the influence of the depth and width of the grating structure on the fluorescence intensity.Rhodamine B(RB)and Rhodamine 6G(R6G)solutions were used to screen and verify different series of substrates to ensure the universality of the substrates.Finally,the optimal substrate is used in the detection of toxic substances crystal violet(CV).
