Tartary buckwheat,a kind of important medicinal and edible crops,has become a research hotspot in recent years because of its antioxidant,hypoglycemic and lipid-lowering effects and anti-atherosclerosis.The content of dietary fiber in Tartary buckwheat is rich.However,the research on the dietary fiber of Tartary buckwheat is only on the content determination,and the activity research is still few.Microbial fermentation is a safe,efficient and economical method to prepare dietary fiber.In this study,Tartary buckwheat dietary fiber was prepared by liquid fermentation of Aspergillus Niger,and its basic physicochemical properties and structure were studied.In order to provide guidance for the development and utilization of Tartary buckwheat dietary fiber(F-DF),a hyperlipidemia mouse model was established to evaluate the regulatory effect of fermented Tartary buckwheat dietary fiber(F-DF)on improving lipid metabolism disorders in mice.The main research contents and results are as follows:(1)Tartary buckwheat dietary fibers were prepared and labeled as UF-DF and F-DF respectively before and after fermentation,and their biological activity indexes were evaluated.The results showed that after liquid fermentation with Aspergillus Niger,the water holding capacity and other capacity of Tartary buckwheat dietary fiber were significantly enhanced,and the glucose adsorption capacity and cholesterol adsorption capacity were improved.Meanwhile,with the increase of total phenol content,the antioxidant activity of DPPH was enhanced.Observation of the two SEM results show that F-DF has a more porous structure,a more fluffy texture and has a better adsorption capacity.The infrared spectrum results showed that both F-DF and UF-DF had typical characteristic absorption peaks of dietary fiber.In conclusion,the fermented Tartary buckwheat dietary fiber had better biological activity.(2)All the mice were randomly divided into five groups after adaptive feeding.Dietetic intervention treatment was conducted for eight consecutive weeks to investigate the effect of F-DF on mice induced by high fat diet.The results showed that after F-DF diet intervention,the weight growth rate of mice induced by high fat diet was slowed down,glucose tolerance was improved,the contents of TC,TG and LDL-C in blood lipids were decreased,and the content of HDL-C was increased,and the dyslipidemic expression of mice induced by high fat was well improved.At the same time,the abnormal expressions of alanine aminotransferase(ALT)and asparate aminotransferase(AST)in liver of mice were reduced by F-DF intervention,and liver steatosis induced by high lipid was improved to a certain extent,and the function of regulating lipid disorders in mice was also improved.(3)The mechanism of lipid-lowering effect of F-DF was initially explored.F-DF can improve liver steatosis and liver fibrosis in mice induced by high fat.F-DF can down-regulate the levels of pro-inflammatory factors(IL-1?,TNF-?)in serum and liver tissue to a certain extent,and inhibit the inflammatory response to a certain extent.F-DF can play a certain lipid-lowering role in mice induced by high fat diet by regulating the NLRP3 inflammatory pathway and reducing the expression of SREBP-1 and FAS protein.(4)The results of high-throughput 16 S r RNA high-throughput sequencing technology showed that F-DF dietary intervention could affect the composition of the intestinal flora of mice,increase the richness of intestinal flora of mice,and regulate the imbalance of intestinal flora.Door and level analysis result,at the same time,the F-DF can by adjusting the high-fat diet-induced the structure of intestinal flora in mice and disorders,raised the bacteroidetes community abundance,cut thick wall fungus door richness,reduce the F/B ratio,improve beneficial bacteria such as lactobacillus abundance in mice,protect the intestinal flora from destruction,adjust and maintain the steady-state,of the intestinal flora in mice intestinal flora imbalance situation.
