Lab on a chip detection technology is also called microfluidic chip detection technology.It is a new type of analysis and detection method that integrates a series of laboratory operations such as sampling,reaction,and analysis on a single chip.It has been developed rapidly since its appearance in the 1990s and become a new research technology in the field of analysis and detection.At the same time,nucleic acid aptamer technology has also been proposed and developed rapidly,becoming a new force in the field of molecular recognition.This project is intended to combine nucleic acid aptamer technology and microfluidic chip detection technology,using chemiluminescence analysis as a detection method to develop a new type of precision detection platform for on-site detection and rapid screening of marine biological toxins and other fields.In this work,our research target is okadaic acid in shellfish which is one of the main bioactive compounds for Diarrheic Shellfish Poisoning(DSP).Rapid,portable detection of Okadaic acid(OA)in shellfish is crucial for practical application in the field of food safety analysis and detection.In this work,a direct competitive aptamer-based chemiluminescent detection of OA in a microfluidic device is demonstrated and used in the real sample analysis.The microfluidic detection device utilizes the OA34 aptamer functionalized polystyrene microspheres as solid phase carriers for capturing targets OA.Horseradish peroxidase(HRP)is utilized to catalyze the luminescence reaction of luminol-H2O2 solution.The coupling of OA to HRP is through the EDC/NHS chemical reaction.Through the direct competition for the limited number of aptamer between O A and HRP-OA,the quantitative detection of OA is realized with a rapid detection of 10 min and a limit of detection(LOD)of 12.4 pg/mL,good resultant correlation coefficient of 0.99938 through the logistics fitting is revealed as well.This detection device in this work enables a rapid,ultrasensitive detection of OA,and demonstrates merits of its field portability and low reagent consumption.The detection device in this work will be deployed to realize the on-site detection and analysis of the marine bio-toxins in the near future.
